浆液性卵巢癌组织miR-105-5p表达变化及临床意义 被引量:1

摘要 目的检测浆液性卵巢癌患者组织中微小RNA(miR)-105-5p表达变化及临床意义。方法选取2018年6月至2021年5月上海交通大学医学院附属瑞金医院舟山分院和上海交通大学医学院附属仁济医院诊治的90例浆液性卵巢癌患者作为研究对象,获取其癌组织为研究组。选取同期诊治的90例卵巢良性肿瘤患者,获取其良性肿瘤组织为对照组。采用实时荧光定量聚合酶链反应(RT-qPCR)检测组织中miR-105-5p水平;分析miR-105-5p与浆液性卵巢癌临床病理特征的关系;采用Spearman分析浆液性卵巢癌癌组织中miR-105-5p与淋巴结转移的相关性;绘制受试者工作特性(ROC)曲线分析癌组织中miR-105-5p对浆液性卵巢癌淋巴结转移的预测价值。结果与对照组比较,研究组miR-105-5p水平降低,差异具有统计学意义(P<0.05)。以浆液性卵巢癌癌组织中miR-105-5p水平<0.72为miR-105-5p低表达组,以癌组织中miR-105-5p水平≥0.72为miR-105-5p高表达组,结果显示miR-105-5p与年龄、肿瘤体积、绝经情况、细胞分化程度、TNM分期无关(P>0.05),与淋巴结转移关系密切(P<0.05)。采用Spearman分析浆液性卵巢癌癌组织中miR-105-5p与淋巴结转移关系,结果显示miR-105-5p与淋巴结转移呈负相关(P<0.05);ROC曲线显示,癌组织中miR-105-5p水平预测浆液性卵巢癌淋巴结转移的ROC曲线下面积为0.753,截断值为0.801,其灵敏度为86.5%,特异度为55.3%。结论浆液性卵巢癌组织中miR-105-5p低表达,与淋巴结转移关系密切,对淋巴结转移有一定诊断价值。 Objective To detect the changes and clinical significance of microRNA(miR)-105-5 p expression in the tissues of patients with serous ovarian cancer. Methods 90 patients with serous ovarian cancer treated in Zhoushan Branch Affiliated to Ruijin Hospital, School of Medicine of Shanghai Jiao Tong University and Renji Hospital Affiliated to Shanghai Jiao Tong University from June 2018 to May 2021 were selected as the study subjects. A total of 90 patients with benign ovarian tumor of the same period were selected as the control group. The level of miR-105-5 p in tissues was detected by real-time fluorescence quantitative polymerase chain reaction(RT-QPCR). The relationship between miR-105-5 p and the clinicopathological characteristics of serous ovarian carcinoma was analyzed. Spearman was used to analyze the correlation between miR-105-5 p and lymph node metastasis in serous ovarian cancer tissues. Receiver operating characteristic(ROC) curves were drawn to analyze the predictive value of miR-105-5 p in serous ovarian cancer lymph node metastasis in cancer tissues. Results Compared with the control group, miR-105-5 p level was decreased in the study group(P<0.05). miR-105-5 p < 0.72 in serous ovarian cancer tissues as the low expression group, and miR-105-5 p≥0.72 in cancer tissues as the high expression group. The results showed that miR-105-5 p was not related with age, tumor volume, menopausal status, degree of cell differentiation and TNM stage(P>0.05), but was closely related with lymph node metastasis(P<0.05). Spearman was used to analyze the relationship between miR-105-5 p and lymph node metastasis in serous ovarian cancer tissues, and the results showed that miR-105-5 p was negatively related with lymph node metastasis(P<0.05). The ROC curve showed that the area under the ROC curve and cut-off value of miR-105-5 p level in cancer tissues for predicting serous ovarian cancer lymph node metastasis were 0.753 and 0.801, respectively, with a sensitivity of 86.5% and specificity of 55.3%. Conclusions The low expression of miR-105-5 p in serous ovarian cancer tissues is closely related to lymph node metastasis, which has certain diagnostic value for lymph node metastasis.
