摘要
目的分析二聚化蛋白(JDP2)、糖皮质激素调节激酶1(SGK1)、糖原合成酶激酶-3β(GSK-3β)在非小细胞肺癌(NSCLC)组织中的表达及相关性。方法选取2018年6月至2020年6月该院收治的76例NSCLC患者纳入肺癌组,病理科均保存有NSCLC组织标本,选择同期76例在该院接受过肺组织活检,并保留有正常肺组织(距肿瘤边缘>5 cm)的患者纳入对照组。采用免疫组化法检测NSCLC及正常肺组织中的JDP2、SGK1、GSK-3β表达情况。结果与对照组相比,肺癌组JDP2、GSK-3β阳性表达率较低,SGK1阳性表达率较高,差异有统计学意义(P<0.05)。与肿瘤最大径<3 cm、无淋巴结转移、TNM分期Ⅰ~Ⅱ期、高分化程度患者相比,肿瘤最大径≥3 cm、有淋巴结转移、TNM分期Ⅲ期、低中分化程度患者JDP2、GSK-3β阳性表达率较低,SGK1阳性表达率较高,差异有统计学意义(P<0.05)。JDP2、SGK1、GSK-3β预测NSCLC发生的受试者工作特征曲线下面积(AUC)分别为0.657、0.695、0.704。结论NSCLC患者JDP2、GSK-3β阳性表达率异常降低,SGK1阳性表达率异常升高,与肿瘤最大径、淋巴结转移情况、TNM分期、分化程度关系密切,且三者呈线性相关,JDP2、SGK1、GSK-3β可能是发生NSCLC的风险预测因子。
Objective To investigate the expression and correlations of dimerization protein 2(JDP2),glucocorticoid regulated kinase 1(SGK1)and glycogen synthase kinase-3β(GSK-3β)in non-small cell lung cancer(NSCLC).Methods A total of 76 NSCLC patients admitted to Zhongda Hospital Affiliated to Southeast University/Nanjing Dachang Hospital from June 2018 to June 2020 were included in lung cancer group,and all NSCLC tissue specimens were preserved in the department of pathology.Other 76 patients who had received lung biopsy during the same period and kept normal lung tissue(>5 cm from the tumor edge)were included in control group.The positive expression of JDP2,SGK1 and GSK-3βin NSCLC and normal lung tissues were detected by immunohistochemistry.Results Compared with those in control group,the positive expression rates of JDP2 and GSK-3βin lung cancer group were lower,and the positive expression rate of SGK1 was higher,the differences were significant(P<0.05).Compared with the patients with tumor maximum diameter<3 cm,no lymph node metastasis,TNM stageⅠ-Ⅱand high differentiation degree,the positive expression rates of JDP2 and GSK-3βin patients with tumor diameter≥3 cm,lymph node metastasis,TNM stageⅢand low middle differentiation degree were lower,and the positive expression rate of SGK1 was higher,the differences were significant(P<0.05).The area under the receiver operating characteristic(AUC)curve of JDP2,SGK1 and GSK-3βwere 0.657,0.695 and 0.704 respectively.Conclusion In patients with NSCLC,JDP2,GSK-3βpositive expression rates decreased,and SGK1 positive expression rate increased,associated with different tumor maximum diameter,lymph node metastasis,TNM stage,differentiation,and JDP2,SGK1,GSK-3βmight be predictors of risk of NSCLC.
