摘要
[目的]运用响应面法优化超声波辅助提取石仙桃总黄酮的工艺,并对其抗氧化活性进行分析。[方法]采用超声辅助提取法在单因素试验的基础上,以提取次数、乙醇浓度、超声功率和料液比为自变量,以石仙桃总黄酮提取率为因变量,运用Box-Behnken设计响应面优化石仙桃总黄酮提取率。[结果]石仙桃总黄酮的超声辅助最佳提取工艺为料液比1∶40(g/m L),超声功率420 W,乙醇浓度75%,石仙桃总黄酮提取率为1.30%,此试验值与响应面模型预测值相差仅为0.69%,在此优化工艺的基础上,以Vc作为阳性对照考察石仙桃总黄酮的体外抗氧化活力,石仙桃总黄酮提取液对DPPH自由基清除能力可达88.35%,对羟自由基可达93.04%。[结论]此优化工艺可为石仙桃总黄酮的提取提供工艺指导,且表明石仙桃总黄酮具有较好的抗氧化活性。
[Objective] Response surface methodology(RSM)was used to optimize the ultrasonic-assisted extraction process of total flavonoids from Pholidota chinensis Lindl,and the antioxidant activity of total flavonoids was analyzed. [Method] The ultrasonic-assisted extraction method was used on the basis of single factor experiment. The extraction times,ethanol concentration,ultrasonic power and solid-liquid ratio were taken as independent variables,and the extraction rate of total flavonoids was taken as dependent variable. The Box-Behnken response surface was designed to optimize the extraction rate of total flavonoids. [Result] The results showed that the optimal ultrasonic-assisted extraction conditions for total flavonoids were solid-liquid ratio of 1 ∶40(g/mL),ultrasonic power of 420 W,ethanol concentration of 75%,and extraction yield of total flavonoids was 1.30%. The difference between the experimental value and the predicted value of response surface model was only 0.69%.Vc was used as positive control to investigate the antioxidant activity of total flavonoids in vitro. The scavenging ability of total flavonoids on DPPH free radical and hydroxyl free radical reached 88.35% and 93.04% respectively. [Conclusion] The optimized technology could provide technical guidance for the extraction of total flavonoid from Pholidota chinensis Lindl,and it shows that the total flavonoids from Pholidota chinensis Lindl has good antioxidant activity.
