胃肠道间质瘤中LMTK3、caspase 3及caspase 7的表达意义

Expression and significance of LMTK3,caspase 3 and caspase 7 in gastrointestinal stromal tumor

目的:探讨胃肠道间质瘤(GIST)中狐猴酪氨酸激酶3(LMTK3)、半胱氨酸天冬氨酸蛋白酶3(caspase 3)及半胱氨酸天冬氨酸蛋白酶7(caspase 7)蛋白的表达与该肿瘤临床病理特征的关系。方法:运用免疫组化EnVision二步法检测102例GIST患者肿瘤组织和相应正常组织中LMTK3、caspase 3及caspase 7蛋白的表达情况,分析LMTK3与caspase 3、caspase 7蛋白表达之间的关系,从而探讨3种蛋白表达与GIST临床病理特征之间的关系。结果:GIST肿瘤组织中LMTK3蛋白表达高于相应正常组织,差异具有统计学意义(P<0.05);LMTK3蛋白表达在不同肿瘤大小、核分裂计数及危险度分组之间比较差异有统计学意义(P<0.05),在不同患者性别、年龄及肿瘤发生部位分组之间差异无统计学意义(P>0.05);肿瘤组织中caspase 3和caspase 7蛋白表达均低于相应正常组织,差异具有统计学意义(P<0.05);caspase 3与caspase 7蛋白表达在患者不同性别、年龄、肿瘤发生部位、肿瘤大小、核分裂像计数及危险度分组之间比较差异均无统计学意义(P>0.05)。GIST肿瘤组织中LMTK3蛋白表达与caspase 3及caspase 7蛋白表达之间无相关性。结论:LMTK3蛋白可能参与GIST发生和发展,其表达水平的升高在一定程度上可预测GIST的恶性潜能。caspase 3和caspase 7蛋白表达水平的降低可能与GIST的发生有关。

Objective:To investigate the relationship between the expression of lemur tyrosine kinase 3(LMTK3),cysteinyl aspartate specific proteinase 3(caspase 3)and cysteinyl aspartate specific proteinase 7(caspase 7)in gastrointestinal stromal tumor(GIST)and the clinicopathological characteristics of GIST.Methods:The EnVision method was used to detect the expression of LMTK3,caspase 3 and caspase 7 in 102 GIST tumor tissues and corresponding normal tissues,and the relationship between LMTK3 protein expression and caspase 3,caspase 7 protein expression was analyzed,so as to explore the relationship between the expression of the three proteins and the clinicopathological characteristics of GIST.Results:The LMTK3 expression was significantly elevated in GIST tumor tissues as compared with corresponding normal tissues(P<0.05);LMTK3 expression was significantly correlated with tumor size,mitotic image and risk degree of GIST(P<0.05),but not with sex,age or tumor location(P>0.05);The expression of caspase 3 and caspase 7 was significantly depressed in GIST tumor tissues as compared with corresponding normal tissues(P<0.05);The expression of caspase 3 and caspase 7 was not related to gender,age,tumor location,tumor size,mitotic image or risk degree of GIST(P>0.05).There was no correlation between LMTK3 protein expression and caspase 3,caspase 7 protein expression in GIST.Conclusion:LMTK3 protein may be involved in the occurrence and development of GIST,and the elevated expression level can predict the malignant potential of GIST to some extent.The depressed expression of caspase 3 and caspase 7 may be related to the occurrence of GIST.
