摘要
目的应用网络药理学方法探讨冬虫夏草预防慢性支气管炎急性发作(AECB)的物质基础及作用机制。方法通过文献检索及TCMSP、Pub-Chem、SwissTargetPrediction在线数据库筛选冬虫夏草的有效成分及靶点预测,从GeneCard数据库获取AECB相关疾病靶点,将药物靶点与疾病靶点进行交集,筛选得到共同作用靶点。将共同作用靶点利用String和Cytoscape3.7.1软件进行蛋白互作网络构建及关键靶点筛选,通过DAVIDE软件进行GO生物功能富集分析和KEGG通路富集分析并通过R version3.6.2软件绘制气泡图。结果本研究共筛选得到符合标准的活性成分7个,药物潜在作用靶点234个,AECB疾病靶点697个,取交集后获得药物与疾病共有核心靶点61个,重点靶点TP53、TNF、MAPK1、EGFR、MAPK8、MAPK14、PTGS2、SRC、AR、NR3C1等,大多与免疫调节、癌症调节及炎症反应相关。GO富集分析得到147条相关条目,主要涉及炎症反应、免疫调控、信号传导、细胞凋亡等生物过程;KEGG富集分析筛选出83条通路,主要包括免疫调节相关通路、感染相关通路、炎症相关通路、癌症相关通路等。结论本研究发现冬虫夏草可以通过调节机体免疫和营造不利于病原微生物的生存环境,发挥防治AECB的作用,为冬虫夏草用于AECB的防治提供理论依据。
Objective:To explore the material basis and mechanism of Cordyceps sinensis in preventing acute at⁃tack of chronic bronchitis(AECB).Methods:The effective components and target prediction of Cordyceps sinen⁃sis were screened through literature retrieval and TCMSP,Pub-Chem and SwissTargetPrediction online databases.AECB related disease targets were obtained from GENECARD database.The drug targets and disease targets were intersected to screen the common targets.String and Cytoscape 3.7.1 software were used to construct protein inter⁃action network and screen key targets,DAVIDE software was used to analyze GO biological function enrichment and KEGG pathway enrichment,and R version 3.6.2 software was used to draw bubble diagram.Results:A total of 7 active ingredients,234 potential drug targets and 697 AECB disease targets were screened in this study.After the intersection,61 core targets were obtained.The key targets TP53,TNF,MAPK1,EGFR,MAPK8,MAPK14,PTGS2,SRC,AR and NR3C1 were mostly related to immune regulation,cancer regulation and inflammatory re⁃sponse.A total of 147 related items were obtained by go enrichment analysis,mainly involving biological processes such as inflammatory response,immune regulation,signal transduction and apoptosis;KEGG enrichment analysis screened 83 pathways,mainly including immune regulation related pathways,infection related pathways,inflamma⁃tion related pathways,cancer related pathways,etc.Conclusion:This study found that Cordyceps sinensis can play a role in the prevention and control of AECB by regulating body immunity and creating a living environment unfa⁃vorable to pathogenic microorganisms,so as to provide a theoretical basis for the prevention and control of AECB by Cordyceps sinensis.
