龙脑樟乙酸乙酯部位的抗炎及抗肿瘤活性研究 被引量:5

摘要 以龙脑樟枝为原料,采用80%的乙醇进行提取,提取液经过乙酸乙酯萃取得到乙酸乙酯萃取相,通过脂多糖(Lipopolysaccharide,LPS)刺激RAW264.7小鼠巨噬细胞建立体外炎症模型,通过Griess法研究龙脑樟乙酸乙酯部位对炎症介质NO生成的影响,MTT法研究其对SH-SY5Y神经瘤细胞和HepG2肝癌细胞增殖的抑制作用。结果表明:在浓度为6.25μg/mL时,龙脑樟乙酸乙酯部位对NO的释放有显著抑制作用,且无明显细胞毒性;在浓度为50~100μg/mL时,乙酸乙酯部位对SH-SY5Y细胞及HepG2细胞的生长均有显著的抑制作用,抑制率均在75%以上。研究表明,龙脑樟乙酸乙酯部位有潜在的抗炎活性及抗癌活性,可为龙脑樟资源的综合开发利用提供理论基础及参考。 In this work,Cinnamomum caraphora were extracted with 80%ethanol,the ethyl acetate(EtOAc)fraction was obtained after ethyl acetate extraction.The effects of EtOAc fraction on the production of inflammatory mediators were determined in the LPS-induced inflammatory responses of RAW 264.7 macrophages.The effect of EtOAc fraction on the production of inflammatory mediator NO was studied by Griess method,and its inhibitory effect on the proliferation of SH-SY5Y neuroma cells and HepG2 hepatoma cells was studied by MTT method.The results showed that the EtOAc fraction of Cinnamomum caraphora significantly inhibited the release of NO and had no obvious cytotoxicity at the concentration of 6.25μg/mL.At the concentration of 50~100μg/mL,the EtOAc fraction significantly inhibited the growth of SH-SY5Y cells and HepG2 cells,and the inhibition rates were more than 75%.It conclude that the EtOAc fraction of Cinnamomum camphor had potential anti-inflammatory and anti-cancer activities,which could provide a theoretical basis and reference for the comprehensive development and utilization of Cinnamomum camphor resources.
