摘要
采用乙醇为提取溶剂,对无刺蜂蜂巢中甾萜类物质的提取工艺进行优化,探究无刺蜂蜂巢乙醇提取物(Ethanol extract of stingless bee honeycomb,EESH)的体外抗氧化活性。采用单因素实验和响应面实验优化EESH的提取工艺;采用香草醛-冰醋酸显色法测定EESH中甾萜类含量,并分析了EESH对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)和2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸[2,2′-azino-bis-3-Ethylbenzothiazoline-6-sulfonic acid,ABTS]自由基清除能力。响应面优化EESH的最佳提取工艺为:乙醇浓度100%、料液比1:20(g/mL)、热浸提温度45℃、热浸提时间150 min,在此条件下甾萜类物质提取得率可达到(36.70±0.64)%;EESH对DPPH和ABTS阳离子自由基的半抑制浓度(IC_(50))分别为(1.350±0.022)mg/mL和(1.984±0.032)mg/mL。上述结果表明无刺蜂蜂巢甾萜类乙醇提取物在体外具有较好的抗氧化活性,为无刺蜂蜂巢高附加值产品的开发提供实验依据。
The extraction technology of steroids and terpenoids in stingless bee honeycomb was optimized with ethanol as the solvent.The antioxidant activities of ethanol extract of stingless bee honeycomb(EESH)were investigated in vitro.Single factor experiment and response surface methodology were used to optimize the extraction process of EESH;steroid and terpenoids in EESH were determined by vanillin-glacial acetic acid color method;the free radical scavenging activities of EESH on 1,1-Diphenyl-2-picrylhydrazyl(DPPH)and 2,2’-Biazo-bis-3-ethylbenzothiazoline-6-sulfonic acid(ABTS)were also analyzed.The optimal extraction conditions of EESH were as follows:ethanol concentration 100%,solidliquid ratio 1:20(g/mL),extraction temperature 45℃,extraction time 150 min.Under these conditions,the yield of steroids and terpenoids could reach(36.70±0.64)%.Half-inhibitory concentrations(IC_(50))of EESH were(1.350±0.022)mg/mL and(1.984±0.032)mg/mL for DPPH· and ABTS^(+),respectively.The ethanol extract of stingless bee honeycomb has good antioxidant activity in vitro,which provides a basis for the development of high value-added products of stingless bee honeycomb.
