摘要
目的探讨maspin、Cyclin-1与胃癌临床分期及血管内皮生长因子(VEGF)的相关性。方法选取2019年1月至2021年5月该院收治的240例胃癌手术患者作为研究对象,收集其癌组织及癌旁组织进行相关指标检测,采用免疫组织化学染色法检测胃癌组织及癌旁组织maspin、Cyclin-1的表达水平,采用酶联免疫吸附试验检测VEGF水平。结果胃癌组织中maspin阳性42例(17.50%),Cyclin-1阳性174例(72.50%);癌旁组织中maspin阳性198例(82.50%),Cyclin-1阳性66例(27.50%)。与胃癌组织比较,癌旁组织中maspin阳性表达较高,Cyclin-1阳性表达较低,差异均有统计学意义(P<0.05)。Ⅰ期胃癌患者maspin阳性21例(26.92%),Cyclin-1阳性36例(46.15%);Ⅱ期胃癌患者maspin阳性12例(21.05%),Cyclin-1阳性48例(84.21%);Ⅲ期胃癌患者maspin阳性9例(17.65%),Cyclin-1阳性42例(82.35%);Ⅳ期胃癌患者maspin阳性0例(0.00%),Cyclin-1阳性48例(88.89%)。各期胃癌患者maspin和Cyclin-1阳性表达情况比较,差异均有统计学意义(P<0.05)。Ⅰ期胃癌患者VEGF表达水平为(62.60±7.22)pg/mL,Ⅱ期胃癌患者为(85.20±15.25)pg/mL,Ⅲ期胃癌患者为(118.30±22.34)pg/mL,Ⅳ期胃癌患者为(140.30±30.56)pg/mL,各期胃癌患者VEGF表达水平比较,差异均有统计学意义(P<0.05)。maspin与VEGF呈负相关(r=-0.365,P<0.05),maspin与Cyclin-1呈负相关(r=-0.298,P<0.05),VEGF与Cyclin-1呈正相关(r=0.386,P<0.05)。结论maspin在胃癌组织中呈低表达,Cyclin-1在胃癌组织中呈高表达,且不同分期胃癌患者maspin、Cyclin-1及VEGF表达水平不同,通过检测maspin、Cyclin-1和VEGF表达水平,对胃癌诊断分期有重要作用。
Objective To investigate the correlation of maspin and Cyclin-1 with clinical stage and vascular endothelial growth factor(VEGF)of gastric cancer.Methods A total of 240 gastric cancer surgery patients admitted to the hospital from January 2019 to May 2021 were selected as the research subjects.The cancer tissues and paracancerous tissues were collected for relevant index detection,and the expression levels of maspin and Cyclin-1 in gastric cancer tissues and paracancerous tissues were detected by immunohistochemical staining.VEGF levels were detected by ELISA.Results In gastric cancer tissue,42 cases(17.50%)were positive for maspin,and 174 cases(72.50%)were positive for Cyclin-1.In paracancerous tissue,198 cases(82.50%)were positive for maspin,and 66 cases(27.50%)were positive for Cyclin-1.Compared with gastric cancer tissue,the positive expression of maspin was higher in paracancerous tissue,and the positive expression of Cyclin-1 was lower,and the differences were statistically significant(P<0.05).In stageⅠgastric cancer,21 patients(26.92%)were maspin positive and 36 patients(46.15%)were Cyclin-1 positive.In stageⅡpatients,12 patients(21.05%)were maspin positive and 48 patients(84.21%)were Cyclin-1 positive.In stageⅢpatients,9 patients(17.65%)were maspin positive and 42 patients(82.35%)were Cyclin-1 positive.In stageⅣpatients,0 cases(0.00%)were maspin positive,and 48 cases(88.89%)were Cyclin-1 positive.There were statistically significant differences in the positive expressions of maspin and Cyclin-1 among patients in different stages(P<0.05).The expression level of VEGF was(62.60±7.22)pg/mL in stageⅠgastric cancer patients,(85.20±15.25)pg/mL in stageⅡgastric cancer patients,(118.30±22.34)pg/mL in stageⅢgastric cancer patients,and(140.30±30.56)pg/mL in stageⅣgastric cancer patients,the differences in the expression levels of VEGF in patients at different stages were statistically significant(P<0.05).Maspin was negatively correlated with VEGF(r=-0.365,P<0.05),maspin was negatively correlated with Cyclin-1(r=-0.298,P<0.05),VEGF was positively correlated with Cyclin-1(r=0.386,P<0.05).Conclusion Maspin was lowly expressed in gastric cancer tissue,while Cyclin-1 was highly expressed in gastric cancer tissue,and the expression levels of maspin,Cyclin-1 and VEGF were different in patients with different stages of gastric cancer.By detecting the expression levels of maspin,Cyclin-1 and VEGF,it plays an important role in the diagnosis and staging of gastric cancer.
