微小RNA-375、erb-B2受体酪氨酸激酶2在卵巢癌组织中的表达及其与病理参数和预后的关系

摘要 目的探讨微小RNA(miRNA)-375、erb-B2受体酪氨酸激酶2(ERBB2)在卵巢癌组织中的表达及其与病理参数和预后的关系。方法选取2016年6月至2018年6月就诊于山东中医药大学附属医院的106例行手术治疗的卵巢癌患者的卵巢癌组织及距离癌组织>5cm的癌旁正常组织作为研究对象。采用实时荧光定量聚合酶链反应(qRT-PCR)法检测卵巢癌及癌旁组织中miR-375、ERBB2的表达,分析卵巢癌组织中miR-375、ERBB2的表达与患者临床病理特征的关系。随访3年,对比不同miR-375、ERBB2表达患者的生存状况,并采用多因素Cox回归分析卵巢癌患者预后的影响因素。结果卵巢癌组织中miR-375的表达低于癌旁组织,ERBB2的表达高于癌旁组织(P<0.001)。Pearson相关系数分析显示,卵巢癌组织中miR-375与ERBB2的表达呈负相关(r=-0.651,P<0.001)。卵巢癌组织中miR-375、ERBB2的表达与分化程度、国际妇产科联盟(FIGO)分期、淋巴结转移有关(P<0.05)。K-M生存曲线显示,miR-375高表达组3年总生存率高于miR-375低表达组,ERBB2低表达组3年总生存率高于ERBB2高表达组(P<0.05)。多因素Cox回归分析显示,FIGO分期、淋巴结转移、ERBB2高表达为卵巢癌患者预后不良的独立风险因素,miR-375高表达为其独立保护因素(P<0.05)。结论卵巢癌组织中miR-375呈低表达,ERBB2呈高表达,二者与分化程度、FIGO分期、淋巴结转移有关,均是患者预后不良的独立影响因素,可作为卵巢癌预后不良的预测指标。 Objective To investigate the expression of microRNA(miRNA)-375 and erb-B2 receptor tyrosine kinase 2(ERBB2)in ovarian cancer and their relationship with pathological parameters and prognosis.Methods The ovarian cancer tissues and the adjacent normal tissues more than 5cm away from the cancer tissues of 106 patients with ovarian cancer who were treated surgically in Affiliated Hospital of Shandong University of Traditional Chinese Medicine from June 2016 to June 2018 were selected as the research objects.Real time fluorescence quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression of miR-375 and ERBB2 of ovarian cancer and adjacent tissues,and analyze the relationship between the expression of miR-375 and ERBB2 in ovarian cancer tissues and the clinicopathological characteristics of patients.After 3 years of follow-up,the survival conditions of patients with different expression of miR-375 and ERBB2 were compared.Multivariate Cox regression analysis was used to analyze the prognostic factors of ovarian cancer.Results The expression of miR-375 in ovarian cancer tissues was lower than that in adjacent tissues,and the expression of ERBB2 was higher than that in adjacent tissues(P<0.001).Pearson correlation coefficient showed that there was a negative correlation between miR-375 and ERBB2 expression in ovarian cancer(r=-0.651,P<0.001).The expression of miR-375 and ERBB2 in ovarian cancer was related to the degree of differentiation,federation international of gynecologists and obstetrics(FIGO)stage and lymph node metastasis(P<0.05).K-M survival curve showed that the 3-year overall survival rate of miR-375 high expression group was higher than that of miR-375 low expression group,and the 3-year overall survival rate of ERBB2 low expression group was higher than that of ERBB2 high expression group(P<0.05).Multivariate Cox regression analysis showed that FIGO stage,lymph node metastasis,high expression of ERBB2 were independent risk factors for poor prognosis in patients with ovarian cancer,and high expression of miR-375 was independent protective factor(P<0.05).Conclusions The expression of miR-375 is low and ERBB2 is high in ovarian cancer tissues,which are related to the degree of differentiation,FIGO stage and lymph node metastasis.Both of them are independent factors affecting the poor prognosis of patients,and can be used as predictive indicators for poor prognosis of ovarian cancer.
