HSF1、sMR、miR-758-3p在肝癌患者中的表达及与临床病理特征的关系

Expression of HSF1,sMR,and miR-758-3p in patients with HCC and their relationship with clinicopathological characteristics

摘要 目的探讨肝癌患者热休克转录因子1(heat shock transcription factor 1,HSF1)、可溶性甘露糖受体(soluble mannose receptor,sMR)、miR-758-3p表达水平变化及其与临床病理特征的关系。方法选取我院进行手术治疗的68例肝癌患者为试验组,另取同时间段来我院就诊的42例慢性肝炎患者为对照组。收集患者临床资料;免疫组织化学检测HSF1水平;酶联免疫吸附测定检测血清sMR水平;RT-qPCR检测miR-758-3p水平;分析HSF1、sMR、miR-758-3p与临床病理特征的关系。结果与对照组比较,试验组HSF1阳性率、血清sMR表达更高,miR-758-3p水平更低(P<0.05);HSF1、sMR、miR-758-3p表达水平与淋巴结转移、微血管侵犯、病理分期、甲胎蛋白(alpha-feto protein,AFP)相关(P<0.05)。HSF1、sMR、miR-758-3p诊断肝癌的曲线下面积为0.947,0.983,0.787,联合HSF1、sMR、miR-758-3p诊断肝癌的曲线下面积为1.000,高于单独诊断。结论肝癌患者HSF1阳性表达显著增加,血清sMR显著升高,miR-758-3p水平显著降低,且HSF1、sMR、miR-758-3p与淋巴结转移、微血管侵犯、病理分期、AFP等临床病理特征密切相关。 Objective To investigate the expression levels of heat shock transcription factor 1(HSF1),soluble mannose receptor(sMR),and miR-758-3p in patients with hepatocellular carcinoma(HCC)and their relationship with clinicopathological characteristics.Methods A total of 68 HCC patients who underwent surgical treatment in our hospital were selected as research subjects,and another 42 patients with chronic hepatitis who presented to our hospital during the same period were selected as controls.Clinical data of patients were collected.HSF1 levels were detected by immunohistochemistry,and serum sMR levels were detected by enzyme-linked immunosorbent assay.RT-qPCR was used to detect miR-758-3p levels.The relationship between HSF1,sMR,miR-758-3p and clinicopathological characteristics was analyzed.Results Compared with the control group,the positive rate of HSF1 and the expression of serum sMR in the experimental group were higher,while the level of miR-758-3p was lower(P<0.05).The expression levels of HSF1,sMR,and miR-758-3p were related to lymph node metastasis,microvascular invasion,pathological stage,and alpha-fetoprotein(AFP)(P<0.05).The area under the receiver operating characteristic(ROC)curve(AUC)of HSF1,sMR and miR-758-3p in the diagnosis of HCC was 0.947,0.983 and 0.787,respectively,and the AUC of combined detection of HSF1,sMR and miR-758-3p in the diagnosis of HCC was 1.000,which was higher than that of single indicator.Conclusion The positive expression of HSF1 and the serum sMR increase significantly,while the level of miR-758-3p decreases significantly in patients with HCC.HSF1,sMR and miR-758-3p are closely related to clinicopathological characteristics,including lymph node metastasis,microvascular invasion,pathological stage and AFP.
