摘要
目的:基于组学和信息学探索山豆根干预血液系统的毒性轮廓。方法:代谢组学和蛋白质组学被用于分析山豆根干预后小鼠血清中差异表达的代谢物和蛋白质。MetaboAnalyst数据库被用于分析这些内源性靶点的联合通路。基于这些靶点,生物信息学数据库被用于筛选山豆根干预血液系统的物质基础。化学信息学数据库被用于预测化学成分的体内分布情况。结果:组学分析表明,经山豆根干预后,小鼠血清中的30个代谢物和83个蛋白质发生差异表达。其中,4个差异表达的代谢物和13个差异表达的蛋白质参与了15个显著调节通路。在成分靶点的介导下,32个成分有可能干预了13个毒性靶点的表达。根据VDss指数,大部分的成分分子趋向于分布在血液中。结论:在毒性剂量下,山豆根有促使细胞凋亡、癌症发展及预后不良、血液疾病、代谢性疾病、炎症等发生的趋势。
Objective:To explore the toxicity profile of Sophorae Tonkinensis radix et rhizoma(ST)in interfering with the blood system based on omics and informatics.Methods:Metabonomics and proteomics were used to analyze the differentially expressed metabolites and proteins in serum after treating with ST.MetaboAnalyst was used for joint pathway analysis.Based on these targets,bioinformatics database was applied to screen the material basis responsible for the intervention of ST on blood system.Cheminformatics database was used to predict the distribution of compounds in vivo.Results:Omics revealed that 30metabolites and 83 proteins were differentially expressed after the treatment with ST,out of which 4 metabolites and 13 proteins were significantly enriched in 15 pathways.Under the mediation of compound targets,32 compounds may interfere with the expression of 13 toxic targets of ST.According to the VDss index,most of the compounds tend to be distributed in the blood.Conclusion:At the toxic dose,ST shows the tendency to promote the occurrence of apoptosis,cancer development and poor prognosis,blood diseases,metabolic diseases,inflammation,etc..
