摘要
目的通过在线数据库挖掘分析RNF111基因在肾透明细胞癌(KIRC)患者组织中的表达、临床意义和生物学功能。方法应用GEPIA、TIMER、UALCAN、HPA、LinkedOmics和DAVID等在线生物信息学工具分析TCGA数据库中RNF111基因在KIRC组织和癌旁组织中表达水平及与患者预后的关系,并对相关蛋白功能和KEGG信号通路进行富集;应用GeneMANIA数据库构建RNF111蛋白-蛋白相互作用网络。结果RNF111在KIRC组织中表达水平显著低于正常肾组织;RNF111在KIRC患者不同病理分期和分级中存在表达差异,且随着病理分期和分级的升高,其表达水平有下调趋势。生存分析表明,RNF111的表达水平与KIRC患者的总体生存率与无病生存率显著相关,RNF111高表达组患者总体生存率和无病生存率明显高于RNF111低表达组患者。GO生物学富集分析结果表明,RNF111与RNA聚合酶Ⅱ启动子的转录延伸、蛋白酶体介导的泛素依赖性蛋白质分解代谢过程及蛋白质多聚泛素化等生物过程有关。KEGG分析结果显示,RNF111表达相关基因与胞吞作用、肾细胞癌及泛素介导的蛋白水解等信号通路相关。结论在KIRC中RNF111的表达与患者的病理分期及预后有相关性,因此RNF111可能成为KIRC的预后标志物及潜在治疗靶点。
Objective To analyze the expression,clinical significance and biological function of RNF111 Gene in kidney renal clear cell carcinoma(KIRC)by mining online databases.Methods Online bioinformatic tools such as GEPIA,TIMER,UALCAN,HPA,LinkedOmics and DAVID were used to analyze,in the database of TCGA,the expression level of RNF111 Gene in KIRC tissues and adjacent tissues and its relationship with the prognosis of patients.The function of related proteins and KEGG signaling pathway were enriched.The RNF111 protein-protein interaction network was constructed using GeneMANIA database.Results The expression level of RNF111 in KIRC tissues was significantly lower than that in normal kidney tissues.There were differences in the expression of RNF111 among patients with different pathological stages and grades of KIRC,with the expression level down-regulated with the increase of pathological stage and grade.Survival analysis showed that the expression of RNF111 was significantly correlated with the overall survival and disease-free survival of KIRC patients.The overall survival rate and disease-free survival rate of patients with high RNF111 expression were significantly higher than those with low RNF111 expression.GO biological enrichment analysis showed that RNF111 was associated with transcriptional extension of RNA polymerase II promoter,as well as proteasome-mediated catabolism of ubiquitin-dependent protein and protein polyubiquitination.KEGG analysis showed that RNF111 expression of related genes were associated with the signaling pathways of endocytosis,renal cell carcinoma and ubiquitin-mediated proteolysis.Conclusion The expression of RNF111 in KIRC is correlated with the pathological stage and prognosis of patients.Therefore,RNF111 may be a prognostic marker and potential therapeutic target of KIRC.
