载补骨脂素微纳米种植体对骨质疏松大鼠骨结合的影响 被引量:3

摘要 目的:研究载补骨脂素微纳米种植体对骨质疏松大鼠骨结合的影响。方法:在钛片表面建立光滑组(ST)、载补骨脂素光滑组(ST-Pso)、微纳米组(MN)、载补骨脂素微纳米组(MN-Pso)、壳聚糖涂覆的载补骨脂素微纳米组(MN-Pso-CHI)5组形貌。扫描电镜观察上述5组表面形貌,紫外可见分光光度计测定ST-Pso、MN-Pso、MN-Pso-CHI组体外补骨脂素释放情况。将上述5组形貌种植体分别植入30只雌性SD骨质疏松大鼠双侧股骨干骺端,28 d后取样进行micro-CT扫描和甲苯胺蓝染色。结果:MN-Pso-CHI组形貌补骨脂素药物释放周期可达15 d,骨体积分数(BV/TV)、骨小梁厚度(Tb.Th)、骨结合率均高于其余4组(P<0.05)。结论:MN-Pso-CHI组形貌种植体补骨脂素可协同微纳米形貌促进早期骨结合,增强种植体的稳定性。 Objective:To study the effects of micro-nanostructure implant loaded with psoralen on the osseointegration in osteoporotic rats.Methods:Titanium sheets were prepared in 5 surface morphologies:smooth(ST),smooth titanium-psoralen(ST-Pso),micro/nano titanium(MN),micro/nano titanium-psoralen(MN-Pso)and micro/nano titanium-psoralen-chitosan(MN-Pso-CHI).The surface morphology was observed by SEM.The release of psoralen of the samples in vitro in the 5 groups was determined by UV-Vis spectrophotometer.The above 5 morphologic implants were implanted into bilateral femoral epiphysis of 30 female SD rats with osteoporosis,and samples were taken 28 days after implantation for micro-CT scanning and toluidine blue staining.Results:The drug release cycle of psoralen in MN-Pso-CHI group was continuously up to 15 days.The bone volume fraction(BV/TV),bone trabecular thickness(Tb.Th)and osseointegration rate in MN-Pso-CHI group were higher than those in the other 4 groups(P<0.05).Conclusion:Psoralen in MN-Pso-CHI titanium implant may promote early osseointegration and enhance the stability of the implants.
