摘要
目的建立桑枝和桑叶的薄层色谱鉴别方法,为其质量控制提供依据。方法基于薄层色谱-静电场诱导喷雾-质谱(TLC-EFISI-MS)联用方法,确定鉴别桑枝和桑叶的特征成分,建立和优化其薄层色谱鉴别方法,并应用于不同批次桑枝和桑叶药材的薄层鉴别。结果通过TLC-EFISI-MS联用鉴定了桑皮苷A为桑枝的特征斑点、紫云英苷为桑叶的特征斑点,以桑皮苷A为指标成分建立了桑枝的特征性薄层鉴别方法,以紫云英苷为指标成分,对《中国药典》中桑叶的薄层鉴别方法进行了优化。对10个不同批次桑枝和桑叶药材进行了薄层鉴别,结果表明在与对照品色谱相应位置上,供试品溶液中均显示相同颜色的斑点且斑点颜色清晰,分离度好。结论通过TLC-EFISI-MS鉴定的桑枝中的桑皮苷A和桑叶中的紫云英苷可较好的区分桑枝和桑叶;建立的薄层鉴别方法操作简便、重复性好,可作为桑枝和桑叶的薄层鉴别方法,为其质量控制提供实验依据。
Objective To develop a TLC identification method for distinguishing Mori Ramulus and Mori Folium.Methods TLC-EFISI-MS was used to identify the characteristic components in Mori Ramulus and Mori Folium,and to establish and optimize their TLC identification methods.Multiple batches of Mori Ramulus and Mori Folium from different sources were identified by the developed TLC method.Results Mulberroside A and astragalin were found as the corresponding specific components of Mori Ramulus and Mori Folium by TLCMS.A characteristic TLC method was thus developed for Mori Ramulus using mulberroside A as a reference substance.The TLC identification method of Mori Folium was improved by using astragalin as a reference substance.The spots in the TLC chromatograms of 10 batches of Mori Ramulus and Mori Folium corresponded to position and colour to the spots in the chromatogram obtained with the reference substance.Conclusion Mori Ramulus and Mori Folium can be easily distinguished by the developed TLC method using mulberroside A and astragalin as reference substances.The developed TLC identification method is easy to operate and reproducible,which can be used as a TLC identification method for quality control of Mori Ramulus and Mori Folium.
