南方菟丝子多成分含量同时测定及聚类热图分析 被引量:1

摘要 本研究建立了南方菟丝子8种成分含量的UPLC测定方法,并进行聚类热图分析。色谱条件:Acquity UPLCBEH C18色谱柱(2.1 mm×50 mm,1.7μm);流动相乙腈-0.1%磷酸水,梯度洗脱;检测波长350 nm;流速为0.3 mL/min;柱温30℃;进样量为1μ L。以各成分含量为分析对象,绘制聚类热图。各成分分离度良好,在线性范围内线性关系良好(R^(2)≥0.9993),平均回收率97.98%~100.21%,RSD为0.85%~1.42%;聚类热图研究结果表明,10批样品可聚为3类,其中1、2、4和6聚为一类;5、7和8聚为一类;3、9和10聚为一类。经方法学验证,所建方法稳定可靠,重复性好,结合聚类热图分析可为南方菟丝子的多成分质量控制与评价提供科学依据。 The UPLC method was established to simultaneously determine the contents of eight constituents in Cuscuta australis.y and cluster heat map analysis was performed.An Acquity UPLC BEH C18 column(2.1 mm×50 mm,1.7 μm)was used,mobile phase was consisted with acetonitrile-0.1%phosphate water by gradient elution,detection wavelength was set at 350 nm,flow rate was 0.3 mL/min,column temperature was set at 30℃,and the injection volume was 1 jjL.The cluster heat map was drawn by taking the content of each component as the analysis object.The eight components in the C.australis all had the good separation and the good linear relationship in linear ranges(R^(2)≥0.9993),the average dosing recovery rates were 97.98%〜100.21%,and the RSD were 0.85%〜1.42%.Cluster heat map analysis showed that 8 batches of samples could be clustered into 3 categories,ofwhich samples 1,2,4 and 6 were clustered into one category,samples 3,9 and 10were clustered into one category,and samples 5,7 and 8 were clustered into one category.The proposed method has been verified by the methodology to be stable and reliable,and has good repeatability,combined with cluster heat map analysis,which could provide a scientific basis for the quality control and evaluation of multi-component of C.australis.
