基于数据挖掘探讨国家专利中药复方治疗多囊卵巢综合征用药规律

Exploration of the medication rules of national patent Chinese herbal compound in the treatment of polycystic ovary syndrome based on data mining

目的:基于数据挖掘对治疗多囊卵巢综合征(polycystic ovary syndrome,PCOS)的国家中药复方专利进行用药规律及配伍分析,为临床选方用药及新药研发提供思路及参考。方法:登录国家知识产权局中国专利公布网站检索治疗PCOS的中药复方专利,运用Excel软件对药物使用频次及性味归经进行统计,运用SPSS Modeler 18.0软件进行Apriori关联规则分析,运用SPSS 26.0软件进行中药系统聚类分析,运用Cytoscape 3.9.1软件构建中药核心复杂网络,并获取各中药的拓扑学属性数据。结果:共纳入97项治疗PCOS的中药复方专利,涉及358味中药,四气以温、平居多,五味以甘、辛居多,归经以肝、脾二经为主;高频中药有香附(38次,39.17%)、当归(33次,34.02%)、菟丝子(30次,30.93%)等,常用对药有“当归-白芍”(支持度18.56%,置信度83.33%)等,常用角药有“熟地黄-淫羊藿-当归”(支持度12.37%,置信度83.33%)等;系统聚类将频次排名前30味中药分为6组,其中含两味及以上中药的聚类有3组;挖掘的新处方为:熟地黄、香附、菟丝子、当归、茯苓、淫羊藿、巴戟天、丹参、益母草、女贞子、枸杞子。结论:治疗PCOS的国家专利中药复方多具有补肾益精、疏肝养血、理气健脾、祛痰化瘀等功效,与PCOS的病因病机具有极强的针对性,其用药规律可为临床治疗PCOS及新药研发提供更多的用药配伍选方思路。

Objective:To analyze the medication rules and compatibility of the national patent Chinese herbal compound in the treatment of polycystic ovary syndrome(PCOS)based on data mining,and provide ideas and references for clinical prescription selection and drug development.Methods:The patents of Chinese herbal compound for the treatment of PCOS were searched on the Chinese Patent Publication website of the China National Intellectural Property Administration.Excel software was used to make statistics on the frequency of drug use and the properties,flavors and meridians of the Chinese herbs.SPSS Modeler 18.0 software was used to analyze association rules of Apriori.SPSS 26.0 was used for systematic cluster analysis of Chinese herbs.The core complex network of Chinese herbs was constructed by Cytoscape 3.9.1,and the topological attribute data of each Chinese herb was obtained.Results:A total of 97 Chines herbal compound patents for the treatment of PCOS were included,involving 358 Chinese herbs.The properties of these Chinese herbs were mainly warm and mild,the flavors were mainly sweet and pungent,and the channel tropism was mainly liver meridian and spleen meridian.The high frequency Chinese herbs included Xiangfu(38 times,39.17%),Danggui(33 times,34.02%),Tusizi(30 times,30.93%)and so on.The common combination of Chinese herbs was Danggui-Baishao(support degeree18.56%,confidence coefficient 83.33%).The common compatibility of three Chinese herbs was“Shudihuang-Yinyanghuo-Danggui”(support degree 12.37%,confidence coefficient 83.33%).The top 30 Chinese herbs with the highest frequency were divided into 6 groups by systematic clustering,among which there were 3 clusters containing 2 or more Chinese herbs.The new prescription was:Shudihuang,Xiangfu,Tusizi,Danggui,Fulin,Yinyanghuo,Bajitian,Danshen,Yimucao,Nvzhenzi and Gouqizi.Conclusion:The national patent Chinese herbal compound used to treat PCOS has the functions of tonifying kidney and nourishing essence,soothing liver and nourishing blood,regulating qi and strengthening spleen,removing phlegm and removing blood stasis,etc.The etiology and pathogenesis of PCOS are highly targeted.The medication rules and compatibility selection can provide more ideas for the clinical treatment of PCOS and the research and development of new drugs.
