多杀菌素发酵罐的发酵放大工艺研究

摘要 为了实现多杀菌素工业化生产,进行了多杀菌素发酵罐放大工艺的研究。探究了溶氧和消泡剂对Saccharopolyspora spinosa YJY-12菌株发酵生产多杀菌素的影响,并对多杀菌素从摇瓶到30 L发酵罐的工艺放大和流加补料发酵进行了探索。摇瓶发酵研究结果显示:Saccharopolyspora spinosa YJY-12菌株发酵生产多杀菌素对溶氧要求较高。以溶氧为依据,在发酵罐间歇发酵过程中,控制溶氧在30%,多杀菌素的发酵效价最大,可达到(2.23±0.06)g·L^(-1)。采用流加补料发酵工艺结果显示:多杀菌素的发酵水平大幅度提高至(3.65±0.08)g·L^(-1)。研究实现了多杀菌素发酵从摇瓶到发酵罐的发酵放大,为多杀菌素的工业化生产提供了有意义的参考。 In order to realize the industrial production of spinosad,the scale-up of the fermentation of spinosad was carried out in this study.The effect of dissolved oxygen and defoamer on the spinosad production of Saccharopolyspora spinosa YJY-12 was determined.Then the scale-up of spinosad fermentation from shake flask to 30 L fermentor,as well as with continuous feeding process were studied.The results of shake flask fermentation showed that Saccharopolyspora spinosa YJY-12 had a higher requirement for dissolved oxygen when producing spinosad.Based on dissolved oxygen,the fermentation level of spinosad was maximum,reach to(2.23±0.06)g·L^(-1) in the batch fermentation process,by controlling the dissolved oxygen content with 30%.Then the fed batch fermentation process was adopt and the fermentation level of spinosad significantly increased to(3.65±0.08)g·L^(-1).Scale-up of spinosad fermentation from shaking flask to fermenter was realized,which provides a meaningful reference for the industrial production of spinosad.
