摘要
基于网络药理学技术探析香砂六君子汤治疗糖尿病胃轻瘫(DGP)的潜在作用机制。通过TCMSP数据库,获取香砂六君子汤相关药物活性成分及蛋白靶点,借助GeneCards、DrugBank、TTD、DisGenet、OMIM数据库挖掘DGP的基因靶点,借助Venny工具获取交集基因,并于String数据库绘建药物与疾病的蛋白相互作用网络。将交集基因借助DAVID数据库进行GO及KEGG通路富集分析。经过筛选得到138个香砂六君子汤的有效活性成分,将其转化为潜在基因靶点得到70个,筛选得到600个疾病相关基因靶点。取两者之间交集得到共同靶点64个,经相互作用分析后高度相关的前5个靶点为血管内皮生长因子A、肿瘤坏死因子、AKT1、白细胞介素-6、前列腺素内过氧化物合酶。KEGG通路富集分析得到信号通路17条,糖尿病并发症AGE-RAGE、癌症、肿瘤坏死因子、PI3K-Akt信号通路等与DGP高度相关。香砂六君子汤通过多成分、多靶点、多通路的相互调控治疗DGP,为其相关潜在作用机制及药理研究提供了参考。
To explore the potential mechanism of action of Xiangsha Liujunzi decoction for the treatment of diabetic gastroparesis(DGP)based on network pharmacology technology.The TCMSP database was used to obtain the active ingredients and protein targets of Xiangsha Liujunzi decoction.GeneCards,DrugBank,TTD,DisGenet,and OMIM databases were used to explore the gene targets of DGP,and the intersecting genes were obtained with the help of Venny tool,and the protein interaction network of the drug and the disease was mapped in String database.The intersected genes were analyzed by DAVID database for GO and KEGG pathway enrichment.After screening,one hundred and thirty-eight active ingredients of Xiangsha Liujunzi decoction were obtained,and seventy of them were converted into potential gene targets,and six hundred disease-associated gene targets were obtained by screening.The intersection of the two components yielded sixty-four common targets,and the top five highly relevant targets after interaction analysis were vascular endothelial growth factor A,tumor necrosis factor,AKT1,interleukin-6,and prostaglandin endoperoxide synthase.KEGG pathway enrichment analysis yielded seventeen signaling pathways,and diabetic complications AGERAGE,cancer,tumor necrosis factor,and PI3K-Akt signaling pathway were highly correlated with DGP.Xiangsha Liujunzi decoction treats DGP through the inter-regulation of multi-components,multi-targets,and multi-pathways,which provides a reference for its related potential mechanism of action and pharmacological research.
