摘要
以新疆纸皮核桃分心木为原料,采用单因素试验考察乙醇体积分数、料液比、提取温度、提取时间对总黄酮提取率的影响,采用响应面试验对工艺进行优化。结果表明:乙醇体积分数56%、料液比1︰43 (g/mL)、提取时间3.6 h、提取温度69℃时,通过热回流法得到的总黄酮提取率为4.131%±0.045%,理论提取率为4.2%,理论值与实际值相差1.64%,误差较小。该工艺操作简单,结果准确,重现性好,具有实际应用价值,为新疆纸皮核桃分心木的深入开发和综合利用奠定基础。
The effect of ethanol concentration,solid-liquid ratio,extraction temperature and extraction time on the extraction rate of total flavonoids was investigated by single factor experment using Xinjiang leatheroid Diaphragma juglandis Fructus as raw material.The response surface methodology was used to optimize this process.The results showed that when the concentration of ethanol was 56%,the ratio of material to liquid was 1:43(g/mL),the extraction time was 3.6 h,and the extraction temperature was 69℃,the extraction rate of total flavonoids was 4.131%+0.045%,and the theoretical extraction rate was 4.2%.The difference between the theoretical value and the actual value was 1.64%,and the error was small.This process had simple operation,accurate results,good reproducibility,and practical application value,laying a foundation for the in-depth development and comprehensive utilization of Xinjiang leatheroid Diaphragma juglandis Fructus.
