异槲皮苷抗腹泻作用的研究

摘要 为了研究异槲皮苷的抗腹泻作用,本试验采用酶水解芦丁法制备异槲皮苷,通过蓖麻油致小鼠腹泻模型、小肠炭墨推进试验和大鼠肠腔积液试验,分析异槲皮苷的抗腹泻作用,并进一步检测其对肠道离子通道蛋白mRNA表达的影响,初步探讨其抗腹泻机制。结果显示,高剂量的异槲皮苷[50 mg/(kg·bw)]可明显延迟蓖麻油诱导的小鼠初次腹泻时间(P<0.01),并减少稀便次数(P<0.05),抑制炭粉在小肠内的推进(P<0.01),并显著减少大鼠的小肠积液(P<0.01);实时荧光定量PCR(qRT-PCR)检测结果显示,与腹泻模型组小鼠相比,异槲皮苷高剂量组小鼠钠/氢交换因子1(NHE1)、钠/氢交换因子2(NHE2)、钠/氢交换因子3(NHE3)、水通道蛋白4(AQP4)和钠/葡萄糖协同转运蛋白1(SGLT1)的mRNA表达显著上调(P<0.05),囊性纤维化跨膜传导调节蛋白(CFTR)的mRNA表达显著下调(P<0.05)。结果表明,异槲皮苷具有明显的抗腹泻作用,为其临床应用提供理论依据。 In order to investigate the anti-diarrheal effect of isoquercitrin,this study employed the enzymatic hydrolysis of rutin method to prepare isoquercitrin,and explored the anti-diarrheal effect of isoquercitrin using castor oil-induced mouse diarrhea model,charcoal meal gastrointestinal propulsion experiments,and rat intestinal fluid accumulation experiments.Additionally,the study examined the impact of isoquercitrin on the mRNA expression of intestinal ion channel proteins and preliminarily delved into its anti-diarrheal mechanism.The results demonstrated that a high-dose of isoquercitrin[50 mg/(kg·bw)]significantly delayed the onset of diarrhea in mice induced by castor oil(P<0.01),reduced the frequency of loose stools(P<0.05),inhibited the propulsion of charcoal through the small intestine(P<0.01),and significantly decreased intestinal fluid in rats(P<0.01).Real-time quantitative PCR(qRT-PCR)results showed that,compared with the mice in the diarrhea model group,the mice in the high-dose isoquercitrin group exhibited significant upregulation of the mRNA expression of sodium/hydrogen exchangers(NHE1,NHE2,NHE3),aquaporin 4(AQP4),and sodium/glucose co-transporter 1(SGLT1)(P<0.05),and significant downregulation of the mRNA expression of cystic fibrosis transmembrane conductance regulator(CFTR)(P<0.05).These findings indicate that isoquercitrin has a notable anti-diarrheal effect,providing a theoretical basis for its clinical application.
