摘要
针对硫酸新霉素药物成分分离不佳的问题,研究在该药物的检测过程中,建立反相高效液相色谱—脉冲安培检测法。该方法的色谱柱选用Thermo Acclaim TM Am G,其内径大小设置为4.6 mm×150 mm。流动相采用2%的三氟乙酸溶液,波形检测的方法使用积分脉冲安培电化学检测。在实验中,硫酸新霉素的主要成分与其他杂质具有良好的分离情况,测出主要成分的检出限与定量限分别为1.75 ng、35 ng。实验方法验证该方法具有较强专属性、高灵敏度和宽线性范围,在实际应用中具有较好的效果。
In the face of the problem of poor separation of neomycin sulfate drug components,a reversed phase high-performance liquid chromatography-pulsed amperometric detection method is established in the detection process of this drug.The chromatographic column of this method is Thermo AcculaimTM AmG,with an inner diameter of 4.6 mm × 150 mm.The mobile phase uses a 2% trifluoroacetic acid solution,and the waveform detection method uses integral pulse amperometric electrochemical detection.In the experiment,the main component of neomycin sulfate has a good separation from other impurities,and the detection limit and quantification limit of the main component are 1.75 ng and 35 ng,respectively.The experimental method verifies that this method has strong specificity,high sensitivity,and a wide linear range,and achieves good results in practical applications.
