摘要
为探究紫锥菊提取物对SD大鼠心肌组织氧化应激的保护作用及相关蛋白表达的影响。选择60只SD大鼠按照随机分组方法分成对照组、不同质量浓度组(10、20、40 mg/kg),每组15只,进行饲养试验。饲养结束后,麻醉并取出心脏,检测心肌组织各生化指标及免疫组织化学染色对心肌组织做病理检测。SD大鼠随着添加紫锥菊提取物浓度增加,SD大鼠氧化应激指标ROS含量和MDA含量呈现逐渐降低的趋势,添加10、20、40 mg/kg紫锥菊提取物组SD大鼠ROS和MDA显著低于对照组(P<0.05);SD大鼠抗氧化指标SOD、GSH-Px和CAT活性呈现逐渐升高的趋势,添加10、20、40 mg/kg紫锥菊提取物组SD大鼠SOD、GSH-Px和CAT显著高于对照组(P<0.05);SD大鼠呼吸链复合体Ⅰ和呼吸链复合体Ⅲ活性呈现逐渐升高的趋势,显著上调线粒体呼吸链复合体Ⅰ、Ⅲ活性(P<0.05);SD大鼠Ca^(2+)-ATP酶呈现逐渐升高的趋势,添加20、40 mg/kg紫锥菊提取物组SD大鼠Ca^(2+)-ATP酶显著高于对照组(P<0.05)。不同试验组间结果显示,SD大鼠添加40 mg/kg紫锥菊提取物效果最佳。低氧适应基因表达结果显示,与对照组相比,添加10、20和40 mg/kg紫锥菊提取物组SD大鼠HIF-1α、EPO和VEGF的mRNA和蛋白表达差异不显著(P>0.05),但有降低的趋势。结果表明,紫锥菊提取物可通过调控SD大鼠心肌组织代谢,增强其抗氧化能力、线粒体功能、能量代谢水平,提高SD大鼠适应能力,且以SD大鼠添加40 mg/kg紫锥菊提取物作用最佳。
This study aims to explore the protective effect of Echinacea extract on oxidative stress in myocardial tissue of SD rats and the expression of related proteins.Sixty SD rats were randomly divided into control group,different concentration groups(10,20,40 mg/kg),15 rats in each group,and feeding experiment was conducted.After feeding,the heart was anesthetized and removed.Biochemical indexes and immunohistochemical staining of myocardial tissue were detected for pathological examination.with the increase of Echinacea extract concentration,the contents of ROS and MDA of oxidative stress indexes in SD rats showed a gradually decreasing trend,and the ROS and MDA of SD rats in 10,20 and 40 mg/kg Echinacea extract groups were significantly lower than those in control group(P<0.05);the activities of SOD,GSH-Px and CAT in SD rats increased gradually.SOD,GSH-Px and CAT in 10,20 and 40 mg/kg Echinacea extract groups were significantly higher than those in control group(P<0.05);the activities of respiratory chain complexⅠandⅢin SD rats were gradually increased,and the activities of mitochondrial respiratory chain complexⅠandⅢwere significantly up-regulated(P<0.05);the Ca^(2+)-ATPase of SD rats increased gradually,and the Ca^(2+)-ATPase of SD rats in 20 and 40 mg/kg Echinacea extract groups was significantly higher than that in control group(P<0.05).The results of different experimental groups showed that SD rats supplemented with 40mg/kg Echinacea extract had the best effect.The results of hypoxia adaptation gene expression showed that compared with the control group,the mRNA and protein expressions of HIF-1α,EPO and VEGF in the 10,20and40mg/kg Echinacea extract groups were not significantly different(P<0.05),but there is a downward trend.Echinacea extract can enhance the antioxidant capacity,mitochondrial function,and energy metabolism of SD rats by regulating the metabolism of myocardial tissue,and improve the adaptive ability of SD rats,and the best effect was achieved by adding 40mg/kg of Echinacea extract to SD rats.
