摘要
目的运用生物信息学分析软件对猴痘病毒A29L蛋白的结构和功能进行预测。方法从NCBI网站上获取猴痘病毒A29L基因及编码A29L蛋白的基本信息,利用NCBI、OFR Finder、ProtParam、ProtScaleon Expasy、SignalP 4.1、TMHMM-2.0、NetPhos 3.1 Sever、SOPMA、SWISS-MODEL、ABCpred、SYFPEITHI、BLAST等生物信息学工具对猴痘病毒A29L蛋白的结构和功能进行预测。结果A29L蛋白是由110个氨基酸组成的亲水性蛋白,含有19种氨基酸,2个开放阅读框。A29L蛋白相对分子质量为12.55924×10^(3),理论等电点5.73,不稳定系数32.28,是稳定蛋白。A29L蛋白存在10个磷酸化位点,无跨膜区、信号肽;A29L蛋白二级结构中α-螺旋占比最高,占57.27%,无规则卷占32.73%;B细胞有11个(score>0.5)抗原表位,CTL细胞有6个(score≥21)抗原表位,Th细胞有10个(score≥20)抗原表位。A29L蛋白有8个同源性较高的蛋白。结论A29L蛋白是具有多个潜在的T、B细胞抗原表位的稳定亲水性蛋白,可作为猴痘治疗和疫苗的候选蛋白。
Objective To predict the structure and function of the monkeypox virus A29Lprotein using bioinformatics analysis software.Methods The basic information of monkeypox virus A29Lgene and encoded A29Lprotein was obtained from NCBI website,and the structure and function of monkeypox virus A29Lprotein were predicted using bioinformatics tools such as NCBI,OFR Finder,ProtParam,ProtScaleon Expasy,SignalP 4.1Sever,TMHMM-2.0,NetPhos 3.1Sever,SOPMA,SWISS-MODEL,ABCpred,SYFPEITHI,and BLAST.Results A29L protein is a hydrophilic protein composed of 110amino acids,containing 19kinds of amino acids and 2open reading frames.The relative molecular mass of A29Lprotein is 12.55924×10^(3),the theoretical isoelectric point is 5.73,and the instability coefficient is 32.28.It is a stable protein.The A29Lprotein has 10phosphorylation sites,no transmembrane region and no signal peptide.The A29Lprotein has the highest proportion ofα-helices(57.27%)and irregular coils(32.73%)in the secondary structure.11(score>0.5)antigenic epitopes were found in B cells,6(score≥21)in CTL cells and 10(score≥20)in Th cells.A29Lprotein has 8proteins with high homology.Conclusion The A29Lprotein is a stable hydrophilic protein with multiple potential T and B cell antigenic epitopes and could be a candidate protein for monkeypox treatment and vaccine.
