摘要
目的 探讨七味白术散对(type 2 diabetes mice, T2DM)小鼠血脂、炎性因子和肠道菌群的影响。方法 将实验动物分为空白组、模型组、阳性组及七味白术散组,采用高脂饲料喂养联合腹腔注射链脲佐菌素造模成功后,给予QWS连续治疗6周,对各实验组小鼠的空腹血糖,血清中血脂生化指标甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)的水平,以及炎性指标小鼠白细胞介素6(IL-6)、小鼠白介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)的水平进行测定。采用16S rDNA高通量测序分析各组小鼠的肠道菌群,探究QWS对T2DM小鼠肠道菌群的影响。结果 与模型组小鼠相比,经QWS治疗后,空腹血糖水平降低,血脂生化指标TC、TG、 LDL-C水平显著降低(P<0.01,P<0.05,P<0.01)。小鼠血清炎性因子TNF-α、IL-6、IL-1β的水平显著降低(P<0.05,P<0.01,P<0.01)。肠道菌群分析结果显示,T2DM小鼠体内厚壁菌门、拟杆菌门、变形菌门、放线菌门和乳杆菌属、HT002、Lachnospiraceae_NK4A136_group、Muribaculaceae_unclassified等的相对丰度呈现差异,经QWS治疗后,QWS组小鼠体内变形菌门、放线菌门、Patescibacteria、梭杆菌门及HT002、Ligilactobacillus、Lachnospiraceae_NK4A136_group等的相对丰度出现不同程度的回调趋势。LEfSe标志菌群分析结果显示,T2DM小鼠的标志性菌群与QWS组明显区别。结论 七味白术散可显著降低T2DM小鼠的空腹血糖及血清中血脂和炎性因子的水平而表现出治疗作用,其机制可能与对T2DM小鼠肠道菌群的调节有关。
Objective To explore the effects of Qiwei Baizhu Powder(七味白术散)on blood lipids,inflammatory factors and gut microbiota in type 2 diabetes(T2DM)mice.Methods The animals were divided into control group,model group,positive group and Qiwei Baizhu Powder group.T2DM mice model was established by high-fat feed feeding combined with intraperitoneal streptozotocin.After 6 weeks treatment,the fasting blood glucose and serum levels of triglycerides(TG),total cholesterol(TC)and low-density lipoprotein cholesterol(LDL-C)as well as inflammatory indicators such as interleukin-6(IL-6),interleukin-1β(IL-1β)and tumor necrosis factorα(TNF-α)were determined.And the gut microbiota of each group was analyzed by 16S rDNA high-throughput sequencing technology.The effect of Qiwei Baizhu Powder on the gut microbiota of T2DM mice was explored.Results Compared with those of the model group mice,after Qiwei Baizhu Powder treatment,the fasting blood glucose level decreased,and the levels of TC,TG and LDL-C significantly decreased(P<0.01,P<0.05,P<0.01).The levels of serum TNF-α,IL-6 and IL-1βsignificantly decreased(P<0.05,P<0.01,P<0.01).The analysis of the intestinal microbiota showed that T2DM mice showed differences in the relative abundance of Firmicutes,Bacteroidota,Proteobacteria,Actinobacteriota and Lactobacillus,HT002,Lachnospiraceae_NK4A136_group and Muribaculaceae_unclassified.After treatment with the Qiwei Baizhu Powder,the relative abundance of Proteobacteria,Actinobacteriota,Patescibacteria,Fusobacteriota,HT002,Ligilactobacillus and Lachnospiraceae_NK4A136_group appeared in Qiwei Baizhu Powder group.The analysis of LEfSe markers showed that the signature flora of T2DM mice was clearly different from the Qiwei Baizhu Powder group.Conclusion Qiwei Baizhu Powder significantly reduced the fasting blood glucose,serum lipids,and inflammatory factors in T2DM mice,demonstrating the therapeutic effects.Its mechanism may be related to the regulation of intestinal microbiota in T2DM mice.
