摘要
目的探讨终末分化诱导的长链非编码RNA(lncRNA TINCR)和成纤维细胞生长因子-1(FGF-1)与肥胖型支气管哮喘患儿病情严重程度及肺功能的关系。方法采用随机抽样法选取我院2021年12月—2023年1月收治的120例支气管哮喘患儿为研究对象,其中肥胖型支气管哮喘患儿60例,非肥胖型支气管哮喘患儿60例;另选取同期于我院进行健康体检的健康儿童120例作为对照组研究对象。又根据肥胖型哮喘患儿的病情严重程度分为轻度组(29例)、中度组(24例)、重度组(7例)。检测所有研究对象的LncRNA TINCR和FGF-1 RNA相对表达水平、嗜酸性粒细胞百分比、第一秒用力呼气容积(FEV1)、最大呼气中段流量(MMEF)等,并分组进行比较分析。结果与非肥胖型支气管哮喘组和对照组相比,肥胖型支气管哮喘组患儿的LncRNA TINCR和FGF-1 RNA相对表达值明显升高(P<0.05);同时,非肥胖型支气管哮喘组患儿的LncRNA TINCR和FGF-1 RNA相对表达值也高于对照组(P<0.05)。肥胖型支气管哮喘组患儿的FEV1%和MMEF显著低于其他2组(P<0.05);另外,非肥胖型支气管哮喘组患儿的FEV1%和MMEF也低于对照组(P<0.05)。在肥胖型支气管哮喘患儿中,重度组的LncRNA TINCR与FGF-1 RNA相对表达水平高于轻度组及中度组(P<0.05)。重度组和中度组的肥胖型支气管哮喘患儿的FEV1%和MMEF都显著低于轻度组(P<0.05)。肥胖型支气管哮喘患儿的LncRNA TINCR、FGF-1与病情严重程度呈正相关(P<0.05),与FEV1%和MMEF呈负相关(P<0.05)。另外,LncRNA TINCR与嗜酸性粒细胞百分比呈正相关(P<0.05)。结论肥胖型支气管哮喘患儿的LncRNA TINCR和FGF-1水平较高,且与病情严重程度及肺功能密切相关。
Objective To explore the correlation between long non-coding RNA terminal differentiation-induced non-coding RNA(LncRNA TINCR)and fibroblast growth factor-1(FGF-1)with the severity and lung function of childhood obesityrelated bronchial asthma.Methods A random sampling method was used to select 120 children with bronchial asthma treated at our hospital from December 2021 to January 2023,including 60 cases of obesity-related bronchial asthma and 60 cases of non-obesity-related bronchial asthma.Additionally,120 healthy children undergoing physical examinations during the same period were selected as the control group.The children with obesity-related asthma were further assigned to the mild(29 cases),moderate(24 cases),and severe(7 cases)groups following the severity of their condition.The relative expression levels of LncRNA TINCR and FGF-1,eosinophil percentage,forced expiratory volume in 1 second(FEV1),and maximal mid-expiratory flow(MMEF)were measured and compared among the groups.Results Compared to the non-obesityrelated asthma group and the control group,the relative expression levels of LncRNA TINCR and FGF-1 RNA were significantly higher in the obesity-related asthma group(P<0.05).Moreover,the expression levels were significantly higher in the non-obesity-related asthma group than in the control group(P<0.05).The FEV1%and MMEF of the obesity-related asthma group were significantly lower than those of the other two groups(P<0.05),and the non-obesity-related asthma group also showed significantly lower values compared to the control group(P<0.05).Within the obesity-related asthma group,the severe group had significantly higher levels of LncRNA TINCR and FGF-1 RNA than the mild and moderate groups(P<0.05).The FEV1%and MMEF in the severe and moderate groups were significantly lower than those in the mild group(P<0.05).LncRNA TINCR and FGF-1 levels in children with obesity-related asthma positively correlated with the severity of the condition(P<0.05)and negatively correlated with FEV1%and MMEF(P<0.05).LncRNA TINCR positively correlated with the percentage of eosinophils(P<0.05).Conclusion The levels of LncRNA TINCR and FGF-1 are elevated in children with obesity-related bronchial asthma and are closely correlated with the severity of the condition and lung function.
