摘要
目的研究反向点杂交(RDB)技术联合血常规检测对β-地贫基因的诊断价值。方法选取2021年1月-2023年3月我院收治的60例β-地中海贫血患者为研究组,并选取同期在我院体检的健康人群60名为对照组,均进行血常规和RDB技术检测。比较两组人群血常规指标[红细胞分布宽度(RDW)、红细胞体积(MCV)、红细胞血红蛋白浓度(MCHC)、红细胞血红蛋白量(MCH)],ROC曲线分析不同检查方法(血常规、RDB、血常规+RDB)诊断β-地贫基因的效能。结果研究组RDW大于对照组,MCV、MCHC、MCH低于对照组(P<0.05);ROC曲线显示,血常规+RDB检查对β-地贫基因检出率为97.86%,高于血常规、RDB单独检测(P<0.05),而血常规与RDB检测对β-地贫基因检出率比较,差异无统计学意义(P>0.05)。结论RDB技术联合血常规检测可提高对β-地贫基因的诊断价值,一定程度减小漏诊、误诊情况,为临床诊治β-地中海贫血提供更可靠的参考,可作为临床筛查诊断β-地贫基因的首选方法。
Objective To study the diagnostic value of reverse dot blot(RDB)combined with blood routine test for β-thalassemia gene.Methods A total of 60 patients with β-thalassemia admitted to our hospital from January 2021 to March 2023 were selected as the study group,and 60 healthy people who underwent physical examination in our hospital during the same period were selected as the control group.The blood routine indexes[red blood cell distribution width(RDW),red blood cell volume(MCV),red blood cell hemoglobin concentration(MCHC),red blood cell hemoglobin(MCH)]were compared between the two groups.ROC curve was used to analyze the efficacy of different examination methods(blood routine,RDB,blood routine+RDB)in the diagnosis of β-thalassemia gene.Results RDW in the study group was higher than that in the control group,MCV,MCHC and MCH were lower than those in the control group(P<0.05).ROC curve showed that the detection rate ofβ-thalassemia gene by blood routine+RDB was 97.86%,which was higher than that of blood routine and RDB alone(P<0.05),while there was no significant difference in the detection rate of β-thalassemia gene between blood routine and RDB(P>0.05).Conclusion RDB technology combined with blood routine test can improve the diagnostic value of β-thalassemia gene,reduce missed diagnosis and misdiagnosis to a certain extent,provide a more reliable reference for clinical diagnosis and treatment of β-thalassemia,and can be used as the preferred method for clinical screening and diagnosis of β-thalassemia gene.
