rhBMP-2和IGF-I联合作用对兔成骨样细胞增殖的影响 被引量:9

摘要 目的:研究人重组骨形成蛋白(recombinant human bone morphogenetic protein,rhBMP),胰岛素样生长因子-1(insulin-like growth factor-1,IGF-I) 单独及联合应用对兔成骨细胞增殖的影响。方法:采用细胞培养技术及噻唑蓝比色法,对2种生长因子对兔成骨样细胞增殖的调节作用进行观察。结果:在培养基中含1%胎牛血清(FBS)条件下,100ng/ml rhBMP-2对兔成骨样细胞增殖作用不显著。IGF-I(5,25,50ng/ml)有一定促增殖作用。两者联合应用各浓度组对细胞增殖有促进作用(P<0.05)。结论:rhBMP-2 和IGF-I联合应用对兔成骨样细胞增殖有协同效应。 Objective:To investigate the effect of recombinant human bone morphogenetic protein(rhBMP-2), insulin-like growth factor -I(IGF-I) on rabbit osteoblastic cell proliferation. Methods:Rabbit bone marrow stromal cells werecultured and induced mineralization to form osteoblast phenotype. The cells were seeded at 1×105/well on 96-well plate.After synchronization, medium was replaced by DMEM containing 1% FBS with various growth fators, concentrationsand combinantions. Cell proliferation was studied by MTT colorimetric method. Result: 100ng/ml rhBMP-2 had noproliferating effect on rabbit osteoblastic cells, while IGF-I had proliferating effect on them at 5-50ng/ml . The synergeticeffects were achieved when they were used in combination. Concluson:These results suggested that rhBMP-2 and IGF-Ihad overlapping effect on rabbit osteoblastic cell proliferation.
