摘要
用流式细胞法测定了 30日龄AA肉鸡的胸腺、法氏囊和脾脏细胞中半胱天冬酶 3(caspase 3)、Bcl 2蛋白的表达及细胞周期和细胞凋亡率。结果 :胸腺、法氏囊、脾脏中G0 /G1 期细胞比例分别是 :96 44%、68 0 9%、92 97% ;G2 /M期细胞比例是 :0 80 %、1 0 4 %、2 36 % ;S期细胞分别为 :2 77%、30 52 %、4 67% ,表明细胞增殖活性的增殖指数 (PI)是 :3 56、31 92、7 0 3。胸腺、法氏囊、脾脏细胞凋亡率分别为 :3 99%、0 32 %、0 2 2 % ;caspase 3表达的阳性细胞率是 :1 4 2 4 %、8 60 %、5 49% ,与细胞凋亡率呈方向一致的变化 ;Bcl 2蛋白表达阳性细胞率依次是 :2 0 59%、30 2 4 %、31 32 % ,与细胞凋亡率呈反向变化。上述结果表明 :鸡免疫器官细胞凋亡是依赖caspase 3介导的细胞凋亡 ,caspase 3和Bcl
We determined the expression of caspase 3 and bcl 2 ,cell cycle and apoptotic rate in thymus,bursa of Fabricius and spleen from 30 day old AA chickens with flow cytometry.The results showed that:(1)The states of cell cycle were that cell ratio of G 0/G 1 phase was 96 44%,68 09%,92 97%,the cell ratio of G 2/M phase was 0 80%,1 40%,and 2 36% and the cell ratio of S phase was 2 77%,30 52%,and 4 67% respectively.Proliferation index(PI)which indicates cell proliferation activity was 3 56,31 92 and 7 03 in thymus,bursa of Fabricius and spleen.(2)Apototic rate was 3 99%,0 32% and 0 22% in thymus,bursa of Fabricius and spleen respectively.(3) Positive cell ratio of caspase 3 expression was 14 24%,8 60%,and 5 49% respectively.The apoptotic changes didnt show statistical difference.(4)The cells of bcl 2 expression were 20 51%,30 24%,31 32% in the above three tissues.Thus bcl 2 expression and apoptosis changed as opposite direction.It indicated that apoptosis in chicken immune organs was dependent on caspase 3.Both caspase 3 and Bcl 2 appear to be two critical regulators for apoptosis of chicken immune organs.
