大蒜素联合5-FU对腺样囊性癌ACC-M裸鼠移植瘤的抑制作用

摘要 目的探讨大蒜素联合5-FU对腺样囊性癌ACC-M裸鼠移植瘤的抑制作用。方法建立ACC-M细胞Balb/c裸鼠肩胛下移植瘤模型30只。造模2周后挑选24只肿瘤大小较均匀的裸鼠随机分为实验组(大蒜素组、5-FU组和大蒜素联合5-FU组)和对照组,各6只。实验组按照体质量0.01 m L/g腹腔注射大蒜素和5-FU浓度均为20μg/m L,对照组注射0.9%PBS液,每两天一次,共3次,末次注射1周后处死动物。测量移植瘤的体积变化,HE染色观察移植瘤的病理学改变。结果移植瘤体积变化比较结果:各实验组移植瘤体积明显缩小,5-FU组体积小于大蒜素组,大蒜素联合5-FU组与大蒜素组、5-FU组比较差异有显著统计学意义(P<0.01);镜下见对照组的癌细胞生长旺盛,异型率高,而各实验组的肿瘤细胞出现不同程度减少,间质增加,大蒜素联合5-FU组最为明显。结论大蒜素和5-FU均能有效抑制ACC-M细胞Balb/c裸鼠移植瘤的生长,且大蒜素联合5-FU的抑制作用明显强于大蒜素或5-FU单独使用。 Objective To investigate the inhibitory effect of allicin combined with 5-fluorouracil(5-FU) on adenoid cystic carcinoma in nude mice models transplanted with ACC-M cells. Methods The nude mice xenograft models were established by subcutaneous injecting human adenoid cystic carcinoma cell line ACC-M into Balb/c nude mice. Twenty-four xenograft model rats were randomly divided into four groups(control group and three experimental groups of allicin group, 5-FU group, and allicin combined with 5-FU group), with 6 rats in each group. The rates in the four groups received intraperitoneal injection of 0.01 m L/g medicine according to the weight of each nude mice, 0.9%phosphate buffer saline(PBS), 20 μg/m L allicin, 20 μg/m L 5-FU, and 20 μg/m L allicin combined with 20 μg/m L 5-FU respectively, once per two days, for three times. The animals were executed after 1 week for measuring the xenograft tumor volume and observing the pathological changing by HE staining. Results The xenograft tumor volume of three experimental groups had narrowed considerably. The tumor volume was significantly decreased in the 5-FU group compared with Allicin group, with statistically significant difference between allicin combined with 5-FU group and allicin group, 5-FU group(P<0.01). Under microscope, the cancer cells of the control group showed vigorous growth and high abnormal rate, while the cancer cells of the three experimental groups showed reduced quantity and increased intercellular substance to different extent, especially in allicin combined with 5-FU group. Conclusion Allicin and 5-FU can both effectively inhibit the growth of nude mice xenograft tumor of adenoid cystic carcinoma. And the inhibitory effects of allicin combined with 5-FU is more significant than either allicin or 5-FU used alone.
