摘要
以2,3,5-三甲吡啶为原料,经N-氧化、硝化、甲氧基取代、乙酰化、水解五步反应合成2-羟甲基-4-甲氧基-3,5- 二甲基吡啶,总收率达69.7%.
2-hydroxymethyl-4-methoxy-3, 5-dimethylpyridine, an intermediate of Omprazole and Esomprazole, was synthesized from 2,3,5-trimethylpyridine by N-oxidation, nitration, and methoxy-lation to give the 4-methoxy-3, 5-dimethylpyridine-N-oxide, which was then treated with acetic anhydride and followed by hydrolysis using dilute hydrochloric acid as hydrolytic reagent to give the title compound. The overall yield reached 69. 7% and the method had the advantage of mild reaction, low cost and waste.
