摘要
目的 :探讨正常脑血管缝隙连接蛋白的分布。方法 :采用RT PCR、免疫组化方法测定多种缝隙连接蛋白在大白兔基底动脉的表达。结果 :(1)RT PCR发现大白兔基底动脉血管组织中至少存在Cx4 5、Cx4 3、Cx4 0和Cx37四种缝隙连接蛋白mRNA表达 ;(2 )免疫组化证实Cx4 5和Cx4 3主要分布于血管内皮 ,平滑肌细胞间未见Cx4 5和Cx4 3的染色。结论 :大白兔基底动脉血管组织存在丰富的缝隙连接蛋白的表达 ,与体内其它弹力血管相比 。
Objective:To investigate the distribution of multiple connexins in the basilar artery of rabbit.Methods:Total RNA was isolated from the basilar arteries and then was used for Revere Transcription Polymerase Chain Reaction (RT PCR).Immunohistochemical examination was performed to determine the distribution of Cx45 and Cx43 in the basilar arteries.Results:The mRNA expressions of at least four different connexins,such as Cx45,Cx43,Cx40,and Cx37,mRNA were found in the basilar arteries.Immunohistochemical analysis demonstrated the present of gap junction proteins Cx45 and Cx43 in the endothelia of the vessels, however neither of them was found in the media of basilar arteries examined.Conclusion:The data indicate multiple connexins are expressed in the basilar arteries of rabbit.The connexin expression patterns are different from other elastic arteries of the body.
