摘要
以黄色短杆菌(Brevibacterium flavum)AG77(AHV^r,TA^r,SG^r)为诱变出发菌株,经亚硝基胍(NTG)诱变处理和选育,获得一株能够积累大量L-精氨酸的菌株AN78(AHV^r,TA^r,SG^r,His^-)。AN78菌株在20L发酵罐中,以葡萄糖为碳源,以玉米浆、硫酸铵等为氮源的培养基中培养4d,产酸率可达61.1g/L,对糖转化率20.2%,与AG77菌株相比较,分别提高了95%和39.3%。发酵液中L-精氨酸的提取收率为71%。
A L-arginine-producing mutant AN78 (AHVr, TAr, SGr, His- ) was derived from Brevibacterium flavum AG77 (AHVr, TAr, SGr) by treatment of N-methyl-N-nitro-N-nitrosoguanidine (NTG)and the fermentative production in 20L fermentor by mutaint AN78 was carried out. It could accumulate 61. 1g/L L-arginine in the medium containing glucose as carbon source and ammonium sulfate as nitrogen source when it was cultured at 30℃ for 4 days. The conversion rate of glucose was 20.2 % . Compared with the parental strain AG77, the yield of L-arginine and conversion rate of mutant AN78 were increased by 95% and 39.3% respectively. The extraction rate of the L-arginine in the fermentated liquor was 71%.
