荧光聚合酶链反应检测严重急性呼吸综合征冠状病毒的方法建立及临床初步应用 被引量:29

摘要 目的 建立荧光聚合酶链反应 (F PCR)检测严重急性呼吸综合征 (SARS)冠状病毒的方法 ,并探讨其临床应用价值。方法 根据公布的SARS冠状病毒基因序列 ,自行设计合成引物、探针 ,应用研制的SARSF PCR诊断试剂盒 ,检测 115份临床漱口液样本。结果 PCR扩增产物经测序证明与SARS冠状病毒基因序列完全一致。 6 7例确诊SARS患者漱口液中 4 9份为阳性 ;18例密切接触者漱口液中 8份为阳性 ;30名健康者漱口液均阴性。结论 建立的F PCR检测SARS冠状病毒方法可成为SARS筛查和早期诊断的一种快速、准确、有效的检测方法。 Objective A fluorescent polymerase chain reaction (F-PCR) method for detecting severe acute respiratory syndrome (SARS) coronavirus was established, and its value for clinical application was evaluated. Methods The primers and the fluorescence-labelled probe were designed and synthesized according to the published sequence of SARS coronavirus genome. A F-PCR diagnosis kit for detecting SARS coronavirus was developed, and 115 clinical gargling liquid samples were detected. Results The sequence of PCR amplified product was completely matched the related sequence of SARS coronavirus gene. 49 in 67 samples from cases of identified SARS patients showed positive. 8 in 18 samples from the cases of closely contact persons showed positive. None of the 30 samples from healthy contrast showed positive. Conclusion F-PCR may become a rapidly, accurate and efficient way for wild-ranged screening and early diagnosis of SARS coronavirus.
