摘要
目的 :研究蕨类植物半边旗提取物 6F对 HL- 60细胞内 c- Myc、Bc1 - 2及增殖细胞核抗原 (PCNA)表达的影响 ,探讨其抗肿瘤作用机制。方法 :异硫氰酸荧光素 (FITC)标记的抗体与细胞反应后在流式细胞仪上检测上述三种蛋白的表达水平。结果 :58~ 2 31 nmo1 / L 6F作用 HL- 60细 1 2 h后 ,c- Myc、Bc1 - 2及 PCNA蛋白表达水平明显下降 ,并呈剂量效应关系。结论 :化合物 6F可下调 HL- 60细胞 c- Myc、Bc1 - 2及 PCNA的表达水平 ,推测这是 6F对 HL - 60细胞具有强烈细胞毒作用并诱导其细胞凋亡的机制之一。
Objective:To study the effects of Compound 6F isolated from Pteris Semipinnata L (PSL)on the expression of c Myc,Bc1 2 and PCNA in HL 60 cells,and to discuss the antitumor mechanisms of 6F Methods:The expression levels of c Myc,Bc1 2 and PCNA were measured by flow cytometry (FCM)after the HL 60 cells were incubated with antibody labelled with fluorescein isothiocyanate(FITC) Results:The expression levels of c Myc,Bc1 2 and PCNA decreased obviously in HL 60 cells treated with 58~231nmo1/L 6F for 12 hours,and the effects appeared in a dose dependence Conclusion:Compound 6F could inhibit the expression of c Myc,Bc1 2 and PCNA in HL 60 cells It is speculated that the cytotoxicity and apoptotic induction of 6F on HL 60 cells related to these effects
