腺病毒介导GDNF的过表达对药物耳毒性损害后的保护作用

摘要 目的 确定腺病毒编码GDNF(glialcellline -derivedneurotrophicfactor)是否能挽救损伤后耳蜗毛细胞的缺失 ,寻求一种治疗药物性耳聋的行之有效的方法。方法  15只杂色豚鼠 ,研究组A注入带有GDNF基因的腺病毒 (Ad -GDNF)而研究组B注入带有GFP基因的腺病毒 (Ad -GFP)。空白对照组C经圆窗注入人工外淋巴液 (AP) ,11d后 15只杂色豚鼠双侧耳蜗行听性脑干反应 (ABR)检查 ,然后处死取材 ,耳蜗标本经硝酸银染色后铺片。结果 研究组听阈值与庆大霉素组和空载体组差异有显著性 ,结果表明Ad -GDNF可能保护庆大霉素损伤后内耳结构和功能。 Objective To determine if glial cell line-derived neurotrophic factor can protect cochlear hair cell from trauma and search for a feasible method in protecting cochlear hair cell from otoxi-induced trauma. Methods Ad-GDNF, Ad-GFP and AP were transferred into inner ears of fifteen guinea pigs through round window respectively. Auditory brain response (ABR) was assessed to these animals preoperatively and postoperatively. On days 11 animals were killed and the cochlear were stained with AgNO 3. Results Animals in the treatment group displayed of otoxicity in the gentamicin or Ad-GFP/gentamicin groups. The data demonstrated protection of the inner ear structure and function against ototoxic insults. Conclusion Adenovirus-mediated glial cell line-derived neurotrophic factor can protect cochlear hair cell from otoxi-induced lesion.
