摘要
目的:探讨内皮素转换酶(ECE)在早期扩张型心肌病(DCM)心室重构中的变化,进一步研究DCM心室重构的发病机制。方法:两肾一夹术制备早期DCM大鼠模型后,通过RT-PCR-半定量法观察ECE mRNA表达水平的变化,并测定ECE活性以了解其蛋白质水平的变化。结果:早期DCM大鼠模型左室ECE mRNA表达水平为:1.25±0.37,ECE活性水平为:(58.3±9.1)pmol/L,二者均较正常对照组明显升高(P<0.05)。结论:早期DCM大鼠模型左室心肌ECE活性及mRNA表达水平与正常对照组相比显著上调,提示ECE可能参与了心室重构的形成。
Objective: To investigate the change of ECE in the early stage of dilated cardiomyopathy (DCM) and further to study the mechanism of ventricular remodeling. Methods: DCM rats models was developed by preparation of hypertension and diabetes rats. To observe the change of ECE mRNA expression by the method of RT-PCR and the protein level of ECE in left ventricle (LV) by measuring ECE activity . Results: ECE mRNA level of LV in DCM rats was higher than that in normal control groups( 1. 25 ± 0. 37 vs. 0. 69 ± 0. 32,P<0. 05). In addition, ECE activity of LV in DCM rats was increased significantly compared with that of normal control groups , (58.319.1 )pmol/L vs. (42. 4 ± 10. 5)pmol/L,P<0. 05. Conclusions: The mRNA expression and activity of ECE in the early DCM was obviously upregulated as compared with those in normal SD rats. This result suggests that ECE may be involved in the formation of ventricular remodeling.
