摘要
目的 通过抗癌药物诱导肿瘤细胞凋亡预测抗肿瘤药物对肿瘤化学治疗的敏感性。方法 用原发性或转移性肺癌组织标本 40份 ,包括组织块 ,胸水、淋巴结。采用 Tunel方法观察抗肿瘤药物诱导肿瘤细胞凋亡。结果 5-Fu诱导肺癌细胞凋亡敏感性分别为高敏 2 0 % ,中敏 5 % ,低敏 5 0 % ,不敏 2 5 % ;CTX分别为 10 % ,0 ,2 5 % ,65 % ;MTX分别为 5 % ,2 0 % ,3 0 % ,45 % ;MMC分别为 10 % ,3 0 % ,3 0 % ,3 0 % ;CDDP分别为 15 % ,3 0 % ,3 0 % ,2 5 % ;CC-NU分别为 15 % ,15 % ,15 % ,5 5 % ;VP-16分别为 15 % ,15 % ,40 % ,3 0 % ;VCR分别为 5 % ,2 5 % ,2 0 % ,5 0 %。结论 用细胞凋亡法预测肿瘤细胞对抗肿瘤药物敏感性经 40份标本检测 ,结果与文献报告的 MTT法 ,3H-Td R,HT-CA相一致 ,但方法简单 ,误差小 ,费时短 ,设备要求低 ,无射线污染 。
Objective Though induction of aplptosis by anticancer drugs.The sensitivity of chemical therapy to cancer cells forecasted. Methods Forty samples are from the tissues,pleural effusion,and lymphonodus.Anticancer drugs induction of apoptosis cancer cells with the method of TUNEL are detected,Results the sensitivity of 5-Fu induction of apoptosis lung cancer cell is expectively,highly sensitive 20%,middle sensitive5%,lowly sensitive 50%,not sensitive25%,CTX is 10%,0,25%,65%.MTX is 5%,20%,30%,45%.MMC is 10%,30%,30%,30%,CDDP is 15%,30%,30%,25%.CCNU is 15%,15%,15%,55%,VP-16 is 15%,15%,40%,30%,VCR is 5%,25%,20%,50%.Conclusions Through observng forty samples,the sensitively of anticancer drugs to the cancer cells are forecasted in apoptosis,The results as well as 3H-TdR,HTCA,which are reported by the method of MTT,but it is simpler ,lest error,taking not much time,less in meed of equipment and no plooution of radiation.So it is worth using widely.
