摘要
目的 :探讨硫酸右旋糖苷 (DS)对人胃癌MKN1细胞株的黏附抑制的机制。方法 :用荧光免疫细胞染色法和共聚焦显微镜观察固定细胞和活细胞在培养盘上的附着过程及形态变化。用Westernblotting法对整合素β 1 蛋白的表达进行分析。结果 :MKN1细胞通过变形 ,形成伪足黏附到培养盘 ,并与其他细胞接触形成单细胞层。免疫染色可见整合素 β 1在细胞膜上有强表达 ,并且形成整合素集落。DS抑制了MKN1细胞的黏附 ,培养48h后仍有部分细胞处于游离状态。DS抑制整合素 β1 的表达并减少已经形成的整合素集落区域。使用DS后 ,黏附细胞整合素β1 蛋白的表达分别减少到非治疗细胞的64 % (130ku)和57 % (110ku) ,游离细胞整合素β 1蛋白的表达分别减少到非治疗细胞的51 % (130ku)和15 % (110ku)。MKN1细胞同时还表现出维持圆形形状的倾向。结论 :DS阻止人胃癌细胞株MKN1的黏附过程与抑制整合素 β1 的表达有关。
Objective:To explore the anti_adhesion mechanism of Dextran Sulf ate(DS)in human gastric canˉcer cell line MKN1in vitro.Methods:The attachment procedure and form change of fixed or living cells were observed by immunofluorescent cytochemistry and confocal laser scanning microscope.Western blot was used to analyze the protein expression of the integrinβ 1 protein.Result:MKN1cells adhered to culture dishes by formˉing filopodia,changed into a flat shape,and adhered to other cells to form monolayer.Integrinβ 1 was intensely expressed in the cell membrane,and formed clusters.DS inhibited the adhesion of MKN1cells,and some cells kept floating in dishes48hours after culture.DS inhibited the impression of integrinβ 1 ,and decreased the clusˉters of integrinβ 1 The expression of integrinβ 1 protein of the adhered cells was decreased to64%(130ku)and57%(110ku)in the untreated cells,and the integrinβ 1 protein expression of the floating cells was decreased to51%(130ku)and15%(110ku)in the untreated cells.Conclusions:The inhibition of the adhesion of MKN1cells by DS was related to the suppression of the expression of integrinβ 1 .
