摘要
目的 初步探讨水通道蛋白 5 (AQP5)在干燥综合征 (SS)小鼠颌下腺中的表达分布。方法 用免疫组化方法和Westernblot分析SS小鼠颌下腺AQP5表达变化特点。结果 对照组AQP5在腺泡的顶膜、侧膜和导管等处均有较强表达 ;实验组AQP5表达减少或消失 ,AQP5在SS残存的腺泡部位 ,顶膜表达减少 ,相应基侧膜的表达增强 ,而且主要分布在导管部位。Westernblot检测显示各组均出现分子量为 2 7× 10 3的条带 ,实验组AQP5的表达量显著减少 (P <0 0 1)。
Objective To investigate the expression distribution of aquaporin 5(AQP 5) in submandibular salivary glands of Sjogren's syndrome (SS) mice. Methods A total of 10 BALB/c mice were injected with pertussis vaccine plus complete Freund's adjuvant (CFA) plus homologous antigen of submandibular gland to enhance the immuological reaction. The other 10 mice, injected only with the same volume of saline, were employed as the control. Samples of submandibular glands, were analyzed histologically, with immunohistochemical method and Western blot quantitative analysis at different time points. Results In the control group, AQP 5 was strongly expressed in the apical membrane, the basolateral membrane, and the secretory duct of the acinar cells. AQP 5 immunoreactivity was predominantly localized at the apical membrane of acinar cells. In the experiment group, however, AQP 5 expression decreased or disappeared; in the remaining acinus, decreased AQP 5 was found in the apical membrane, but enhanced expression of AQP 5 was found in the corresponding basolateral membrane, mainly distributed in the basal membrane and duct cells. Western blot analysis showed that there was a 27×10 3 band in both groups, and AQP5 expression in the experiment group decreased significantly ( P <0 01). Conclusion The significant decrease of AQP 5 expression and abnormal distribution of AQP 5 in salivary gland acini in SS mice may be associated with dry mouth.
