γ-射线及大肠杆菌诱导蓖麻蚕产生抗菌物质的研究 被引量:7

摘要 1.用γ-射线辐照和用大肠杆菌处理蓖麻蚕五龄幼虫和蚕蛹均能诱导血淋巴产生抗菌物质,两种诱导源诱导所得活性物质的抗菌活性相似。2.研究了五龄幼虫期和蛹期诱导产生抗菌物质的动力学,发现五龄幼虫在饷食后第2—4天诱导活力最高,产生抗菌物质的持续时间亦较长;蛹期则从化蛹当天至第4天之间诱导活力较高,并可持续15天左右,高峰期一般在诱导后2天至4天之间。3.对诱导后的蓖麻蚕血淋巴进行了电泳测活和葡聚糖凝胶初步分离,发现不论幼虫期或蛹期至少可得三个活性组分,其中既有类似于P_5的大分子抗菌物质,也有类似于P_(9A)和P_(9B)的抗菌多肽;并首次发现一种分子量约70000—75000道尔顿的新抗菌蛋白。 Both the larvae in fifth instar and the pupae of Philosdmia cynthia ricini can be induced to produce antibacterial substances in the haemolymph by irradiation with γ-rays or injection with Escherichia colt. The antibacterial activities of the active substances induced by these two factors are similar. It was found that the most competent stage of. the insect for the induction was two to four days after starting the feeding in fifth larval instar and the duration of the competence was about three to six days, ,In the pupal, stage the most competent stage was from the first to the fourth day after the last larval moult which could last for about 15 days. In general the peak of the immunological reaction was in the period of two to four days after the treatment.The antibacterial substances in. the haemolymph of the larvae and' pupae induced by injection with E. coli was studied by electrophoresis (PAGE) and isolated with sephadex and se-phrose. It was, found that at least three active components could be obtained and they included a p5-like large molecule and a p98-like polypeptide in both the larval and pupal stages. A. new antibacterial protein with MW about 70,000-75,000 dalton was found.
