摘要
目的建立尿中吗啡的简便快速、灵敏可靠的GC/NPD分析方法.方法样品尿加内标烯丙吗啡,酶或酸催化水解,氯仿-异丙醇(9:1)液液提取或GDX403树脂固相提取,BSA衍生化,HP-5柱和氮磷检测器进行分析.结果提取率62%~85%,检出限1.2~3.1ng/ml,线性范围20~2000ng/ml,回收率(97%~99%)±(6%~9%)(Mean±cv,N=5).结论方法适合于实际案件中尿样的检验.
Objective Sensitive, reliable, simple and rapid methods for the determination of morphine in urine by gas chromatography and nitrogen-pnosphorus detection were developed. Methods The urine samples were added to nalorphine (I. S. ) and hydrolyzed with glucuronidase or acid as catalyst. The hy-drolysates were liquid-liquid extracted with chloroform-isopropanol (9: 1) as extraction solvent at pH8. 9 condition or were solid-phase extracted with GDX403 porous resin as sorbent and with chloroform as eluent at pH8. 9 on. The extracts were derivatized with BSA. The derivatives were chromatographed on HP-5 column and detected by nitrogen-phorphorus detector. Results The extraction efficiencies were 61% to 85% , the limits of detection were 1. 2 ng/mL to 3. 1 ng/mL, the linearity were good in the range 20 ~ 2000ng/mL, the recoveries were 97% to 99% and the CV of it were 6% to 9%. Conclusion The methods reported in this paper are proper for actual case examination.
