妊娠高血压综合征亚甲基四氢叶酸还原酶基因和血管紧张素转换酶基因多态性研究 被引量:6

Study on MTHFR gene and ACE gene polymorphisms in pregnancy-induced hypertension

摘要 目的 探讨N5,N10 亚甲基四氢叶酸还原酶 (methyenetetrahydrofolatereductase ,MTH FR)基因和血管紧张素转换酶 (angiotensin convertionenzyme,ACE)基因多态性与妊娠高血压综合征的关系。 方法 应用多聚酶链反应 限制性内切酶片段长度多态性 (PCR RFLP)技术检测 6 2例妊娠高血压综合征 (妊高征组 )患者及 12 0例正常妊娠 (对照组 )的ACE和MTHFR基因多态性。 结果 妊高征组MTHFR基因T/T基因型频率 ( 2 7% )显著高于对照组 ( 15 % ) (P <0 .0 5 ) ,T等位基因频率 ( 5 2 % )也显著高于对照组 ( 39% ) (P <0 .0 1) ,妊高征组ACE基因缺失型纯合子 (DD)频率( 4 0 % )显著高于对照组的 ( 10 % ) (P <0 .0 1) ,缺失型 (D型 )等位基因频率 ( 6 0 % )显著高于对照组的( 2 9% ) (P <0 .0 1)。MTHFR基因T等位基因与ACE基因D等位基因之间存在协同作用。 结论 MTHFR基因和ACE基因多态与妊高征的发病有关 。 Objective To investigate the methyenetetrahydrofolate (MTHFR) gene and angiotensin convertion enzyme (ACE) gene polymorphisms in pregnancy induced hypertension(PIH). Methods The MTHFR and ACE gene genotypes were determined in 62 pregnancy induced hypertension patients and 120 normal pregnant women by PCR RFLP. Results The frequencies of T allele(0.52) and the T/T genotype(27%) of MTHFR gene in PIH group were markedly higher than those in the control group(39% and 15% P<0.05,P<0.01 respectively) ; The frequencies of D allele(60%) and the D/D genotype(40%) of ACE gene in PIH group were markedly higher than those in the contral group(29% and 10% P<0.05, P <0.01 respectively). The individuals with TT+DD, TT+ID or CT+DD were 1.4 , 3.1, 2.7 times susceptible to PIH than the controls. Conclusion Genetic variations in MTHFR and ACE gene could be involved in the risk of PIH.There may be a co effective action between polymorphisms of ACE gene and MTHFR gene in PIH.
