甲状旁腺素相关肽诱导骨髓细胞生成破骨细胞及对骨吸收的影响 被引量:6

摘要 目的:建立甲状旁腺素相关肽(PTHrP)诱导的破骨细胞培养系统,并观察其特点。方法:分离小鼠骨髓细胞后,用0,20,50,100ng的PTHrP刺激,第6天时观察生成的破骨细胞数目。用PTHrP50ng刺激,观察培养后4,6,8,10,14d时破骨细胞生成数目。结果:破骨细胞数目随PTHrP剂量的增高而增加。培养后4d有破骨细胞形成,6~8d破骨细胞数达到高峰,14d时几乎消失。结论:PTHrP可诱导破骨细胞形成,破骨细胞形成数目与甲状旁腺素相关肽呈剂量依赖,破骨细胞寿命少于两周。 AIM:To create cell culture system so as to examine the characteristics of osteoclast formation induced by parathyroid hormone related peptide (PTHrP). METHODS:When bone marrow cells were stimulated for 6 days by 0,20, 50,100 ng PTHrP separately, seporately or bone marrow cells were stimulated for 4,6,8,10,14 days by 50 ng PTHrP,the number of osteoclasts was studied. RESULTS:The number of osteoclasts increased as PTHrP dose increased. Osteoclasts formed 4 days after culture,and reached the maximum 6-8 days later, while nearly disappeared after 14 day culture. CONCLUSION:PTHrP can induce osteoclast formation which is associated with the number of ostoclasts in a close dependent manner.Life span of osteoclast is less than 2 weeks.
