摘要
由于肌苷生产菌枯草杆菌JSIM - 10 19,腺嘌呤缺陷 ,但黄嘌呤并不缺陷。以鸟苷产生菌枯草杆菌JSIM -G - 5 18为供体菌 ,得到编码IMP脱氢酶的guaA基因。并用氯氨苄抗性基因插入guaA基因 ,使之不产生活性IMP脱氢酶 ,从而可制备黄嘌呤缺陷的新菌株 ,从而可以制备肌苷产量高于JSIM - 10
The bacterium Bacillus Subtilis JSIM-1019 need adenine,but it was not of auxotroph xanthine.GuaA gene was got which coding IMP dehydrogenase from Bacillus subtilis JSIM-G-518 which can accumulate guanosine.Then were guaA gene inserted in with the gene resisting CM.So the living IMP dehydrogenase could be clean away,and the new strain was got which was of auxotroph xanthine,and it could accumulate inosine more than JSIM-1019.
