摘要
目的进一步研究FTY720的降低外周血淋巴细胞的效果和剂量及用药时间的关系。方法给予FTY72010mg/(kg.d),于给药前、第1次给药后2、4、8、24、48h采血计数外周血淋巴细胞数目;给予不同剂量的FTY720(0.3、1、3、10mg/(kg.d))和生理盐水,给药后24h,采血计数外周血淋巴细胞数目。结果给药前、第1次给药后2、4、8、24、48h外周血平均淋巴细胞数目分别为(9.575±1.1121)×109、(6.525±1.0175)×109、(3.512±1.0171)×109、(3.576±1.0046)×109、(3.513±1.0582)×109、(3.500±0.9606)×109/L;生理盐水和不同剂量的FTY720(0.3、1、3、10mg/kg)组的外周血平均淋巴细胞数目分别为(9.475±1.1121)×109、(7.825±1.3092)×109、(6.250±1.0821)×109、(5.100±0.9501)×109、(3.325±0.9977)×109/L。结论FTY720诱导外周血淋巴细胞数目减少有时间和剂量依赖性。给予FTY720(10mg/(kg.d))能够明显降低外周血淋巴细胞数目,并于用药后2h下降明显,用药后4h下降到谷值,4h以后维持于较低水平。外周血淋巴细胞数目和FTY720的剂量成直线关系,随FTY720剂量的增加而下降。
Objective To probe into the relationship between effect of lymphopenia and time and dose of administration of FTY720.Methods Giving FTY720 10mg/(kg·d) 2 days, take blood specimen at the time piont before and 2、4、8、24、48 h after administration to calculate the sum of lymphocyte in periblood. Giving FTY720 (0.3,1,3,10mg/(kg·d)) and 0.9% NS, take blood specimen 24 h after administration to calculate the sum of lymphocyte in periblood.Results Mean sum of lymphocyte in periblood are (9.575±1.1121)×109、(6.525±1.0175)×109、(3.512±1.0171)×109、(3.576±1.0046)×109、(3.513±1.0582)×109、(3.500±0.9606)×109/L aspectively at the time piont of before and 2、4、8、24、48 h after administration of FTY720.Mean sum of lymphocyte in periblood are (9.475±1.1121)×109、(7.825±1.3092)×109、(6.250±1.0821)×109、(5.100±0.9501)×109、(3.325±0.9977)×109/L aspectively in the group of 0.9% NS and different dose of FTY720 (0.3、1、3、10 mg/(kg·d)).Conclusion FTY720, as a new class of immunosuppressant, could induce lymphopenia and show a time and dose dependent mechanism. The mean sum of lmyphocyte in periblood decrease obviousely 2 h after administration, reachs nadir 4 h after administration, and maintain at low level stabely. The mean sum of lmyphocyte shows a linar relationship with dose of FTY720 and decrease with addition of dose of FTY720.
