实时定量PCR分析CML患者胸腺近期输出功能 被引量:10

Recent Thymic Output Function in Patients with Chronic Myelogenous Leukemia by Real Time PCR Analysis

摘要 目的 了解慢性粒细胞白血病 (CML)患者CD3+ 、CD4 + 和CD8+ 细胞中T细胞受体重排删除DNA环 (TRECs)的含量 ,从而推测CML患者的胸腺近期输出功能。方法 利用实时定量PCR(Taq Man)方法检测 8例CML患者外周血单个核细胞、3例CML患者CD4 + 和CD8+ 细胞中TRECs的水平。并根据外周血中CD3阳性率计算CD3+ 细胞中TRECs水平。 9例正常人外周血作为对照。结果 CML患者中TRECs含量为 0 .1± 0 .2 2 / 10 0 0T细胞 ,明显低于正常人TRECs水平 (6 .84± 4 .71/ 10 0 0T细胞 ,P <0 .0 1)。其中 5例患者外周血单个核细胞中未能检测到TRECs ,经分选CD4 + 和CD8+ 细胞后 ,2例可以检测到低水平的TRECs。结论 本研究首先报道了CML患者TRECs水平情况 。 Objective To quantitative analysis the T cell receptor excision DNA circles (TRECs) in CD3 +, CD4 + or CD8 + T cells, evaluating the thymic recent output function in patients with chronic myelogenous leukemia (CML).Methods Quantitative detection of T cell receptor excision DNA circles (TRECs) in DNA of peripheral blood mononuclear cells from 8 cases with CML, CD4 + or CD8 + cells from 3 cases with CML were preformed by real time PCR (TaqMan) analysis. And the TRECs number was related to the number of T cells by determination of the number of CD3 positive cells. 9 normal individuals served as controls.Results A dramatic reduction of TRECs values in patients with CML was showed. The mean value of TRECs was 0.1 ± 0.22 copies /1 000 T cells in CML patients and 6.84 ± 4.71 copies /1 000 T cells in normal individuals, respectively ( P < 0.01 ). In five out of eight CML cases no TRECs copies could be detected in peripheral blood, and lower value of TRECs could be identified in selected CD4 + or CD8 + cells.Conclusion This is, to our knowledge, the first description of TRECs level in CML patients. These results show a prominent reduction of TREC levels in CML.
