摘要
通过有限稀释法由粉纹夜蛾(Trichoplusia ni)Tn 5 B_1细胞系中分离出多个细胞株,经筛选获得一个理想的克隆株Tn 5 B_(1-4),该克隆株的特性不同于原细胞系Tn_5B_1。该克隆株的细胞形态一致,细胞群体倍增时间较原细胞株短.而且细胞活力强,特别是在维持粉纹夜蛾单粒包埋核型多角体病毒(Tn SNPV)的复制能力方面高于原细胞系。另外经生物测定看出,在该克隆株中增殖的病毒毒力与虫体内增殖的病毒无显著差异。
A clonal strain Tn5B_(1-4)was successfully isolated from the established Trichoplusia ni cell line Tn5B, by the method of finite dilution. The growth characteristics of the clonal strain were different from the parent cell line in its morphology, kinetics of cells, cell doubling time and the relative ability to support replication of TnSNPV. The bioassay of TnSNPV from Trichoplusia ni larvae and cloned strain was conducted for comparing their virulence.
