甲醛对V79细胞DNA、RNA合成影响的体外实验研究 被引量:3

摘要 目的 研究甲醛对V79细胞DNA和RNA的损伤作用 ,探讨甲醛遗传毒性的分子机制。方法 中国仓鼠肺细胞 (V79)分别暴露于浓度为 75 ,15 0 ,30 0 ,6 0 0 μmol/L的甲醛中 ,1h、2h、4h ,采用3 H -胸腺嘧啶核苷 ( 3 H -TdR)、14 C -尿嘧啶核苷 ( 14 C UR)掺入技术检测甲醛致V79细胞DNA和RNA损伤情况。结果 所有 4个浓度组甲醛均明显影响细胞DNA和RNA的合成 ,3 H TdR和14 C UR掺入计数每分衰变数 (dpm值 )显著低于对照组 (P <0 .0 1) ,并有明显的剂量 -效应关系 (P <0 .0 1)。细胞暴露在甲醛中 4h ,各浓度组3 H TdR和14 C UR掺入dpm值显著低于暴露于 1h和 2h各浓度组的dpm值 (P <0 .0 1)。 结论 甲醛可以影响V79细胞DNA和RNA合成 。 Objective To explore genotoxic mechanism of formaldehyde(FA) and the effect of formaldehyde(FA) the damage of DNA and RNA of V79 Chinese hamster cells. Methods V79 Chinese hamster cells were exposed to formaldehyde at different concentration of 75、150、300、600μmol/L and incubated for 1、2、4h respectively.The effect of FA on DNA and RNA in V79 Chinese hamster cells was detected by using the assay of radioactive compounds ( 3H-thymidine and 14 C-Uridine)incorporation. Results The decrease of 3H-thymidine and 14 C-Uridine incorporation (dpm) of DNA and RNA in all FA-treated cells and a significant dose-dependent decrease were observed ( P <0.01).The incorporation(dpm) of 75、150、300、600 μmol/L FA-treated cells at 4h was significantly lower than that of ones at 1 h and 2 h ( P <0.01). Conclusion The results suggest that FA may affect DNA and RNA synthesis in V79 Chinese hamster cells and indicate that FA can contribute to FA-induced carcinogenesis.
