摘要
目的 研究甲醛对中国仓鼠肺细胞 (CHL细胞 )DNA和RNA的损伤作用 ,探讨甲醛遗传毒性的分子机制。方法 CHL细胞分别暴露于浓度为 75 ,15 0 ,30 0 ,6 0 0 μmol/L的甲醛中 1,2 ,4h ,采用3 H-胸腺嘧啶 (3 H TdR)、 14 C -尿嘧啶 (14 C UR)掺入技术 ,检测甲醛对CHL细胞的DNA和RNA合成的影响。结果 所有 4个浓度组甲醛均明显影响CHL细胞DNA和RNA的合成 ,3 H TdR和14 C UR掺入计数dpm值显著低于对照组 (P <0 0 1) ,并有明显的剂量 -反应关系 (P <0 0 1)。CHL细胞暴露在甲醛中 4h ,各浓度组3 H TdR和14 C UR掺入dpm值显著低于暴露于 1h和 2h的各浓度组 (P <0 0 1)。结论 甲醛可以影响CHL细胞DNA和RNA合成 ,这对进一步研究甲醛遗传毒性的分子机制有重要意义。
Objective To study the damage of DNA and RNA in CHL Chinese hamster cells induced by formaldehyde and explore its genotoxic mechanism.Methods CHL Chinese hamster cells were exposed to formaldehyde(FA)at final concentration of 75,150,300,600 μmol/L and incubated for 1,2,4 h respectively.The influence of FA on DNA and RNA in CHL Chinese hamster cells was investigated by using the assay of radioactive compounds( 3H-thymidine and 14 C-Uridine)incorporation.Results The decrease of 3H-thymidine and 14 C-Uridine incorporation of DNA and RNA in all FA-treated cells and a significant dose-dependent decrease were observed(P<0.01).The incorporation of 75,150,300,600 μmol/L FA-treated at 4 h was significantly lower than that of ones at 1 h and 2 h( P<0.01 ).Conclusions The results suggest that FA may affect DNA and RNA synthesis in CHL Chinese hamster cells and indicate that FA can contribute to FA induced carcinogenesis.
