摘要
本文合成了3种不同载体蛋白的雌二醇抗原,研究了载体蛋白免疫原性对雌二醇抗体产生的影响。应用酶联免疫吸附分析法测定了血清中雌二醇,浓度范围为5~160ng/ml(1.8×10^(-8)~5.9×10^(-7)mol/L).RSD%小于10.0%,检测限为1.97 ng/ml,相当于98.5pg/孔(按空白平均值的二倍标准偏差计算)。另外,本文还建立了一种用考马斯亮蓝G-250测定半抗原与蛋白结合比的方法,提出了一种新的亲和层析法纯化雌二醇抗血清的方法。
We synthesized estradiol antigens with three different carrier proteins, studied the influence of carrier protein's immunogenicity to produce estradiol antibody. Estradiol in serum was determined by the enzyme-linked immunosorbent assay, the linear range of concentration was 5-160 ng/ml, or 1.8×10^(-8)-5.9×10^(-7) mol/L with the relative standard deviation of less 10%. The detection limit was 1. 97 ng/ml, equivalent to 98. 5 pg per well (2 × standard deviation of mean value from zero standard). In addition, we established a new method to estimate the ratio of hapten to carrier protein with coomassic brilliant blue G-250 and purity estradiol antiserum by the affinity chromatography.
